摘要
为了阐明牡丹形态多样性与遗传背景及亲缘关系的远近,本研究利用EST-SSR分子标记技术,从66对EST-SSR引物中筛选出条带清晰、多态性好的15条引物对57份紫斑牡丹品种的遗传多样性进行分析。结果表明15对EST-SSR标记引物在57份材料中共扩增出65个多态性位点,每对引物平均扩增出4.333个等位基因,变化范围为2~7条;多态信息含量(polymorphism information content, PIC)在0.132~0.836之间,平均为0.618;这表明受试的57份紫斑牡丹品种具有丰富的遗传多样性,所选用的15对EST-SSR引物标记具有较高的多态性。57份紫斑牡丹品种间遗传相似系数范围在0.521 2~0.800 0之间,表明这些品种之间遗传背景相似而又存在差异。UPGMA聚类结果表明,在遗传相似系数为0.587 0处可将57份品种分为4类,第Ⅰ类为‘四川’牡丹1个品种,第Ⅱ类为‘黄云’1个品种,第Ⅲ类为4个品种(2个花色),第Ⅳ类为其他不同花色紫斑牡丹51个品种。同时反映出遗传聚类组划分与花色、花型的划分无明显相关性。研究结果为紫斑牡丹种质资源的整理、新品种选育工作提供科学依据。
In order to elucidate the morphological diversity, genetic background and genetic relationship of Paeonia rockii, EST-SSR molecular marker technology was used to detect the genetic diversity of 57 Paeonia rockii cultivars by fifteen primers with clear band and good polymorphism selected from 66 pairs of EST-SSR primers. The results showed that, among the 57 varieties, 15 pairs of primers marked by EST-SSR amplified 65 polymorphic sites. Each pair of primers amplified 4.333 alleles in average which ranged from 2 to 7. Allelic polymorphism information content (PIC) ranged from 0.132 to 0.836 with an average of 0.618. This result showed that the 57 varieties of Paeonia rockii were rich in genetic diversity, and 15 EST-SSR primers selected had high polymorphism. The genetic similarity coefficient of 57 varieties of Paeonia rockii was between 0.521 2-0.800 0, indicating that these cultivars had similar genetic background but with difference. Clustering analysis based on the UPGMA method revealed that these 57 Paeonia rockii cultivars could be divided into four groups at similarities coefficient of 0.587 0. The group I included one cultivar of P. rockii ' Sichuan'. The group Ⅱ included one cultivar of P. rockii 'Huangyun'. The group Ⅲ included 4 varieties (2 colors). Group IV included 51 Paeonia rockii cultivars with different flower. The results showed that there was no significant correlation between group division of genetic clustering and division of color and flower type. The study would provide scientific basis for the management of Poeonia rockii germplasm resources and breeding of new varieties.
