产褐藻胶裂解酶菌株HB12274的鉴定和发酵优化 被引量:5

摘要 鉴定一株从红树林土壤中分离的产褐藻胶裂解酶菌株HB12274,并优化其培养条件使之产生高活性的胞外褐藻胶裂解酶。依据形态学、生理生化特性和16S rDNA序列分析对菌株HB12274进行鉴定;设计单因素和正交试验优化菌株HB12274产胞外褐藻胶裂解酶的培养条件。菌株HB12274鉴定为解淀粉芽孢杆菌植物亚种Bacillus amyloliquefaciens subsp. plantarum;菌株HB12274产褐藻胶裂解酶的最适发酵条件为:褐藻酸钠5.0 g/L、蛋白胨6.0 g/L、Na Cl 15.0 g/L、CaCl21.0 g/L、初始pH6.0,30℃发酵培养42 h。在最优培养条件下,菌株HB12274最大酶活性可达721.2 U/mL,与优化前酶活力相比提高了2.95倍。本研究获得菌株Bacillus amyloliquefaciens HB12274的最适产酶条件,为该菌的进一步研究应用提供参考。 To identify an alginate lyase-producing strain HB12274 isolated from mangrove forest and optimize the fermentation conditions for alginate lyase production. Strain HB12274 was identified on the basis of the morphology, biochemical and physiological analyses, as well as phylogenetic analysis of 16S rDNA sequence. The fermentation conditions for alginate lyase production by strain HB 12274 were optimized using the one-factor-at-a- time and orthogonal method. Strain HB 12274 was identified as Bacillus amyloliquefaciens subsp, planmrum, and the optimal fermentation conditions were obtained as follows: sodium alginate 5.0 g/L, peptone 6.0 g/L, NaCl 15.0 g/L, CaCl2 1.0 g/L, original pH 6.0, 30℃ for 42 h. Under the optimized fermentation conditions, the highest alginate lyase activity of 721.2 U/mL was achieved, which was increased 2.95-fold compared with the control. The optimum alginate lyase-producing conditions of Bacillus amyloliquefaciens subsp, plantarum 12274 was obtained, which provided the basis for research and application of this strain and alginate lyase.
