失笑散不同溶剂提取物对ox-LDL诱导HUVECs损伤的保护作用 被引量:7

Protective Effects of Different Solvent Extracts of Shixiao Powder on ox-LDL-induced Human Umbilical Vein Endothelial Cell Injury

摘要 目的:观察失笑散各溶剂提取物对氧化型低密度脂蛋白(oxidized low density lipoprotein,ox-LDL)诱导的人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)损伤的保护作用,探讨失笑散保护血管内皮细胞的有效提取物及其可能作用机制。方法:体外培养HUVECs,并制备失笑散石油醚、乙酸乙酯、甲醇提取物。采用ox-LDL诱导制备HUVECs损伤模型,并采用失笑散各溶剂提取物干预损伤细胞。采用MTT比色法测定各组细胞增殖情况,试剂盒检测各组细胞上清液中超氧化物歧化酶(superoxide orgotein dismutase,SOD)的活力、谷胱甘肽-过氧化物酶(glutathione peroxidase,GSH-px)活力、TBA法测定丙二醛(maleic dialdehyde assay,MDA)含量。结果:25 mg·L^(-1)ox-LDL可抑制血管内皮细胞活性。与模型组比较,失笑散石油醚提取物(25.00 mg·L^(-1),12.50 mg·L^(-1))及甲醇提取物(25.00 mg·L^(-1),12.50 mg·L^(-1))可显著提高细胞活力(P<0.01,P<0.05),显著降低MDA含量,提高SOD、GSH-px活性(P<0.01,P<0.05),且失笑散甲醇提取物在25 mg·L^(-1)质量浓度下,与失笑散石油醚提取物比较,在MDA含量、SOD活力的指标上表现出更有效的保护作用(P<0.05)。结论:失笑散石油醚及甲醇提取物可显著降低ox-LDL诱导的HUVECs损伤,为失笑散保护血管内皮细胞的有效提取物,其机制可能与其降低MDA含量和提高SOD、GSH-px活性有关,通过调控氧化损伤机制保护血管内皮细胞。 Objective : To study the protective effects of solvent extracts of Shixiao Powder on oxidized low density lipoprotein ( ox- LDL) induced human umbilical vein endothelial cell (HUVECs) injury and its possible mechanism of action. Methods: HUVECs were cultured in vitro, and human umbilical vein endothelial cell injury model was induced by 25 mg · L-1 ox-LDL. The prolifera- tion and the vitality of superoxide orgotein dismutase (SOD) and glutathione peroxidase (GSH-px) in the supernatant of each group were determined by MTY colorimetry. , TBA method to determine the content of maleic dialdehyde (MDA). Results : 25 mg· L^-1 ox-LDL can inhibit the vascular endothelial cell activity. Compared with that of the model group, petroleum ether extract ( 25.00 mg · L^- 1,12.50 mg· L^-1 ) and methanol extract (25.00 mg· L^-1 ) significantly increased the cell viability ( P 〈 0.01,P 〈 O. 05 ), significantly decreased MDA content, increased SOD, GSH-px activity ( P 〈 0.01, P 〈 O. 05 ). At the dose of 25 mg · L-1, compared with the petroleum ether extract group, methanol extract showed more effective protection on the indicators of MDA content and SOD activity (P 〈0.05). Conclusion : The oil and methanol extract of Shixiao Powder can significantly reduce the ox-LDL-induced cell injury of HUVECs being an effective extract of vascular endothelial cells,which may be related to the re- duction of MDA content and the increase of SOD and GSH-px activities. Besides, it can protect vascular endothelial cells by regula- ting the mechanism of oxidative damage.
