摘要
目的观察二陈汤对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠模型肺组织中水通道蛋白5(aquaporin 5,AQP5)表达的影响及意义。方法采用气道滴入脂多糖加烟熏诱导制备COPD大鼠模型。依据肺功能和组织病理评价COPD大鼠模型是否成功。随机将大鼠分为正常组、模型组、二陈汤低、中、高剂量组。正常组、模型组给予等量生理盐水灌胃,二陈汤低、中、高剂量组分别给予二陈汤2.5、5、10 g·kg^(-1)灌胃,连续14 d。检测各组大鼠肺功能,RTqPCR检测AQP5 mRNA表达,蛋白免疫印迹法(Western blotting)检测肺组织AQP5蛋白定量表达,免疫组化检测AQP5在肺组织中的定位表达。结果与正常组相比,模型组大鼠肺功能显著下降(P<0.05),其组织结构基本符合COPD病理特征,模型组肺组织AQP5 mRNA (P<0.01)和蛋白(P<0.05)表达显著减弱。与模型组相比,二陈汤中、高剂量组肺功能显著改善,肺组织AQP5的mRNA (P<0.01)和蛋白(P<0.05)表达均显著增强。结论二陈汤改善肺功能,可能与上调AQP5基因表达有关。
Objective To observe the effect of Erchen decoction on aquaporin 5 (AQP5) expression in puhnonary tissues of rats with chronic obstructive puhnonary disease (COPD) . Methods COPD model was induced by cigarette smoking combined with lipopolyrsaccharide. The success of the COPD rat model was evaluated on the basis of lung function tests. The rats were randomly divided into normal control group, model group, Erchen decoction group (low, middle and high) . The normal group and model group were given physiological saline by intragastric, while other groups were given corresponding herbal drugs intragastrically (2.5, 5, 10 g/(kg'd)) for 14 days. RT-qPCR and Western blot were used to examine the mRNA and protein expression of AQP5 in puhnonary tissues. Results Comparing with normal group, the lung function test showed that the structure of FEV1/FVC〈0.70 of model group was basically consistent with the pathological features of COPD. The expressions of AQP5 mRNA (P〈0.01) and protein decreased significantly in the model group (P〈0.05) . Comparing with the model group, FEV1/FVC increased significantly (P〈0.05) , and expression of AQP5 mRNA and protein significantly increased in Erchen decoction of 5 g/kg and 10 g/kg groups. Conclusion The mechanism of action of Erchen decoction for regulating COPD airway mucus hypersecretion may be related to upregulation of the expression of and AQP5 gene.
