摘要
为获得海州常山愈伤组织和不定芽,以海州常山叶片作为外植体,研究了不同消毒方法、培养基种类、培养条件和植物生长调节剂配方对海州常山叶片愈伤组织诱导和不定芽再生的影响。研究结果表明:海州常山叶片灭菌最适方法为75%酒精处理30 s,再用0.1%Hg Cl2溶液处理5 min;暗培养有助于愈伤组织的诱导;愈伤组织诱导的最适培养基为MS+6-BA 0.50 mg·L^(-1)+NAA 0.10 mg·L^(-1),诱导率最高达66.67%;愈伤组织最适增殖培养基为MS+6-BA 0.50 mg·L^(-1)+NAA 0.05 mg·L^(-1),愈伤组织平均大小达21.76 mm;最佳分化培养基为MS+6-BA 0.50 mg·L^(-1)+NAA 0.01 mg·L^(-1),分化率为13.33%。
For establishing the system of callus induction and adventitious shoot regeneration from leaves of Clerodendrum trichotomum , the experiment was conducted with the young leaves in vitro to study the effects of disinfection methods, basic culture media, culture conditions and plant growth regulators on the callus induction and adventitious shoot regeneration. The most appropriate method of surface sterilization on leaves was treating with 75% alcohol for 30 s, and then 0.1% Hgcl 2 for 5 min. The dark environment was conducive to the callus induction. The optimal medium for callus induction from leaves was MS containing 6-BA 0.50 mg·L^-1 and NAA 0.10 mg·L^-1 with a maximum induction rate of 66.67%. The optimal medium for callus proliferation was MS containing 6-BA 0.50 mg·L^-1 and NAA 0.05 mg·L^-1 with the average callus size of 21.76 mm, and the optimum medium for adventitious shoot differentiation was MS containing 6-BA 0.50 mg·L^-1 and NAA 0.01 mg·L^-1 with a 13.33% differentiation rate.
