摘要
由于人口的增长和老龄化,肿瘤发病率逐年增加。有研究表明,信号通路的激活和细胞因子的参与均可影响肿瘤的发展。脂氧合酶(LOX)作为脂质过氧化酶可催化多元不饱和脂肪酸形成生物脂参与人体多方面生理过程的调节,包括肿瘤。15-LOX在癌症的发生、发展中也发挥促进或抑制的作用,其中一个亚型15-LOX-1被认为是前列腺癌的致癌因子,在结肠癌中发挥抑制作用。而另一个亚型15-LOX-2在抑制前列腺癌的同时还能促进肾癌的发展。目前15-LOX在癌症中的确切发病机制仍不明确。
The occurrence of tumor is increasing because of the growth and aging of population. A number of studies show that the activation of signaling pathways and the involvement of cytokines all influence the development of tumors.Lipoxygenase( LOX),as a member of lipid-peroxidizing enzymes,catalyzes polyunsaturated fatty acids toform biological lipids to participate in many aspects of physiological progresses,including various neoplasms. The suppressing or promoting effects of 15-LOXs in the occurrence and development of tumors have been demonstrated. For example,one of the subtypes,15-LOX-1 is considered to be a carcinogen of prostate cancer and plays an inhibitory role in colon cancer. Another subtype,15-LOX-2,can inhibit prostate cancer but contributes to the progression of renal cancer. The exact pathogenesis of 15-LOX in diverse cancers is still unclear.
