摘要
目的建立舒筋活血颗粒的质量评价方法。方法采用UHPLC-MS-MS(LC-MS)法,对舒筋活血颗粒中2种活性成分葛根素、黄芪甲苷进行同时测定,采用Agilent poroshell C_(18)色谱柱(2.1 mm×100 mm,2.7μm),以0.2%甲酸-乙腈(体积比40∶60)为流动相,4 min内等度洗脱,共测定10批次供试品。结果葛根素、黄芪甲苷在4 min内全部洗脱,2种成分在10~1 000 ng/mL下线性关系良好(r^2=0.999 2和0.999 4),已测定的10批次供试品中葛根素的平均质量分数为3.42 mg/g,RSD为8.12%;黄芪甲苷的平均质量分数为0.028 mg/g,RSD为19.63%。结论本方法准确、快速、高效,为以该制剂为代表的复方中药制剂的质量评价提供了参考。
Objective To establish quality evaluation method for Shujin Huoxue Keli. Methods UHPLCMS-MS was used for simultaneous determination of puerarin and astragaloside A,two active components of Shujin Huoxue Keli. An Agilent poroshell C18 column( 2.1 mm× 100 mm,2.7 μm) was used with a mobile phase containing 0. 2% formic acid and acetonitrile( 40 ∶ 60), the separation was carried out in isocratic mode within 4 min,and 10 batches of Keli were analyzed. Results Puerarin and astragaloside A were eluted within 4 min,with good linearity( 0.999 2 and 0.999 4,respectively) in the concentration range of 10-1 000 ng/mL. For 10 batches of Keli,the mean content of puerarin was 3.42 mg/g( in dried granules),RSD was 8.12%; the mean content of astragaloside A was 0.028 mg/g,RSD was 19.63%. Conclusions The study proposed an accurate,fast and effective method for simultaneous determination of main active components in Shujin Huoxue Keli,which provides a reference for the evaluation of complex TMC formula.
