摘要
为探讨红花檵木带节茎段的离体快繁技术,以当年生红花檵木幼嫩枝条茎段为外植体,研究外源激素及培养基类型对其丛芽诱导、丛芽增殖、生根培养的影响和不同基质对组培苗驯化移栽的影响。结果表明:红花檵木的丛芽诱导率最佳培养基配方为MS+6-BA 1.4mg/L+NAA 0.1mg/L,其出芽数和诱导率分别为90个和90%;丛芽增殖培养基最佳为MS+6-BA1.3mg/L+NAA0.2mg/L,丛芽数和增殖系数分别为170株和3.4,MS+6-BA2.0mg/L+NAA 0.2mg/L处理下丛芽数和增殖系数虽然最高,但丛芽玻璃化数及玻璃化率也最高,分别到达18个和36%;红花檵木的组培苗生根诱导最佳培养基配方为1/2MS+NAA 0.4mg/L+IAA 3mg/L,其生根数和生根率最高,分别为45条和90%,且此时生根量多、根长且健壮;驯化移栽最适宜的基质配比为珍珠岩︰蛭石︰泥炭=1︰1︰4,成活株数为42株,成活率达到84%,幼苗叶片舒展,生长健壮,新根萌发数量多。试验结果表明,红花檵木幼嫩带节茎段作外植体能够实现其离体快繁。
The main factors of bud induction,buds proliferation,rooting culture and transplanting of exogenous hormones of Loropetalum chinense var. rubrum were studied with the tender stem segments as explants in Loropetalum chinense var. rubrum tissue culture,to provide data support for the establishment of stem segment with node micro-propagation system.The results showed that:the young stem segment of Loropetalum chinense var. rubrum with node as explants could achieve micropropagation.The best medium for bud induction was MS+6-BA 1.4 mg/L +NAA 0.1mg/L,with the germination number and induction rate 90 and 90% respectively;the best medium for buds proliferation was MS+6-BA 1.3mg/L+ NAA 0.2mg/L,which the bud number and proliferation index were 170 and 3.4;under the treatment of MS+6-BA 2.0mg/L+ NAA 0.2mg/L,the bud number and proliferation times were the highest,but the hyperhydric bud number and hyperhydric rate were also the highest,reaching 18 and 36% respectively;the optimum rooting medium was 1/2MS+NAA 0.4mg/L+ IAA 3mg/L,with the highest rooting rate and root number,respectively 45 and 90%,and with the most root numbers,long and strong root;most suitable substrate ratios for Loropetalum chinense var. rubrum acclimatization and transplantation were perlite:vermiculite:peat =1︰1︰4,which the survival number reached 42 strains,the survival rate 84%,with stretch seedling leaves,strong growth,and most new root germination numbers.
