摘要
目的探讨益生菌联合制剂对便秘的改善作用及相关机制。方法30只小鼠随机分为正常组、便秘组和益生菌组,每组10只。便秘组和益生菌组给予洛哌丁胺皮下注射5mg·kg-1,每天2次,连续7天,建立小鼠便秘模型。正常组给予等体积生理盐水。造模成功后,益生菌组给予益生菌联合制剂10mg·kg-1灌胃,其余组给予等体积生理盐水,连续干预14天。检测各组小鼠的粪便指标和小肠转运率;苏木素-伊红染色观察各组小鼠结肠组织变化;免疫印迹法检测各组小鼠结肠组织中B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl相关蛋白(Box)蛋白含量。结果与正常组相比,便秘组粪便指标和小肠转运率显著下降,结肠组织中Bcl-2/Bax蛋白表达比例下调,差异有统计学意义(P〈0.05);黏膜层杯状细胞减少,腺体减少,大量炎性细胞浸润。与便秘组相比,益生菌组粪便指标和小肠转运率显著改善,结肠组织中Bcl-2/Bax蛋白表达比例上调,差异有统计学意义(P〈0.05);结肠黏膜损伤减轻,腺体和杯状细胞修复,炎性细胞浸润明显改善。结论长期使用益生菌制剂可显著改善小鼠的便秘症状,有效改善结肠组织损伤,这种改善作用可能与上调Bcl-2抑制Box的表达从而抑制细胞凋亡有关。
Objective To explore the effects and mechanism of probiotics in the treatment of constipation. Methods A total of 30 mice were randomly divided into three groups : a normal group, a constipation group, and a probiotic group (n = 10). The constipation group and probiotic group were intraperitoneally injected with 5 mg· kg-1 of loperamide, twice per day for 7 days for establishment a mouse constipation model. The normal group was given the equal volume of normal saline. After successful modeling, the probiotic group was given 10 mg·kg-1 probiotic by gavage, while the normal and constipation groups received an equal volume of normal saline for 14 consecutive days. Then, the fecal parameters and small intestinal transition rate were detected. The changes of the colon tissues were observed by H - E staining. The levels of Bcl - 2 and Bax were determined by Western blotting. Results Compared with the normal group, the con- stipation group showed remarkable decreases in fecal parameters, small intestinal transition rate and the levels of Bcl - 2/ Bax in colon tissues ( P 〈 0.05 ). Compared with the constipation group, fecal parameters, small intestinal transition rate and the levels of Bcl - 2/Bax in colon tissues were significantly increased in the probiotic group (P 〈 0.05). Meanwhile the histological parameters were also improved in the probiotic groups. Conclusions Probiotics can significantly relieve the symptoms of constipation in mice, which may be associated with inhibiting apoptosis through Bcl - 2 up - regulation and Bax down- regulation.
