五味子乙素调控PI3K/Akt信号通路介导大肠癌细胞SW116凋亡 被引量:7

Effects of schisandrin B in colon cancer cell line apoptosis through regulating PI3K/Akt signaling pathway

摘要 目的观察五味子乙素对大肠癌细胞SW116凋亡及PI3K/Akt信号通路相关蛋白表达的影响。方法 SW116细胞培养后共分为4组,分别为空白对照组、五味子乙素不同浓度(10、20、40μg/m L)组,对各组细胞进行相应的干预。采用CCK8法检测五味子乙素对SW116细胞株的增殖抑制作用,流式细胞术检测五味子乙素对SW116细胞凋亡的影响,免疫印迹技术分析五味子乙素处理的SW116细胞中凋亡蛋白及PI3K/Akt的表达水平。结果 CCK8实验结果显示,五味子乙素能明显抑制SW116细胞株的增殖;流式细胞仪检测表明,五味子乙素可以诱导SW116细胞株的凋亡;Western blot结果显示,五味子乙素可以诱导促凋亡蛋白Bax的表达,抑制抗凋亡蛋白Bcl-2,还可明显抑制PI3K、p-Akt蛋白的表达。结论五味子乙素可以抑制大肠癌细胞增殖,其诱导肿瘤细胞的凋亡作用可能是通过抑制PI3K/Akt信号通路活化来实现的。 Objective To investigate the effects of schisandrin B on SW116 cell apoptosis of colon cancer cells( SW116) and on the expression of the related proteins in PI3 K/Akt signaling pathway. Methods The SW116 cells were divided into four groups after cell culture,including blank control group and schisandrin B groups at different concentration( 10,20 and 40 μg/m L),and the cells in each group were intervened accordingly. The proliferation inhibition of schisandrin B on SW116 cells was analyzed with CCK8 assay; the influence of schisandrin B on SW116 cells apoptosis was determined with FCM; the expression levels of apoptotic proteins,PI3 K and p-Akt in SW116 cells treated with schisandrin B were assessed with Western blot method. Results CCK8 assay displayed that schisandrin B could inhibit the proliferation of SW116 cell line. Flow cytometry displayed that schisandrin B could stimulate the SW116 cells apoptosis. Western blot showed that the expression levels of Bcl-2,PI3 K and p-Akt were greatly decreased,while the levels of Bax were significantly increased. Conclusion Schisandrin B can inhibit the proliferation of SW116,and the cell apoptosis induced by it may be through inhibiting the activation of PI3 K/Akt signaling pathway.
