中医药对大肠癌多药耐药作用及机制 被引量:13

Effects and Mechanism of Chinese Herbs Against Multidrug Resistance in Colorectal Carcinoma

摘要 多药耐药是导致大肠癌化疗失败的主要原因之一。研究表明肠胃清、至真方、左金丸等中药复方,白花蛇舌草、徐长卿等中药,以及苦参碱、姜黄素、吴茱萸碱等中药单体可以逆转大肠癌多药耐药,其机制涉及多药耐药的多个方面,包括:(1)抑制膜转运蛋白过度表达(如P-gp、MRP等),提高胞内药物浓度;(2)抑制NF-κB信号转导;(3)抑制抗凋亡基因表达,如Survivin;(4)抑制Hedgehog信号转导;(5)抑制MAPK信号转导;(6)抑制上皮-间质转化等。此外,中药还对耐药大肠癌细胞具有直接的抗癌作用。合理应用这些研究成果将有助于提高大肠癌的临床治疗疗效。 Multidrug resistance is one of the major causes of chemotherapy failure in colorectal cancer. Studies have shown that Changweiqing,Zhizhenfang,Zuojinwan and other Chinese herb formulae,Hedyotis diffusa,Cynanchum paniculatum and other Chinese herbs,as well as matrine,curcumin,evodiamine and other herbal compounds can reverse multidrug resistance in colorectal cancer. Chinese herbs reverse multidrug resistance in colorectal carcinoma through multiple mechanisms,including:(1)inhibition of membrane transporters overexpression( such as P-gp and MRP) to improve intracellular drug concentration;(2)inhibition of NF-κB signal transduction;(3)inhibition of anti-apoptotic gene expression,such as Survivin;(4)inhibition of Hedgehog signal transduction;(5)inhibition of MAPK signal transduction;(6)inhibition of epithelial-mesenchymal transformation. In addition,Chinese herbs also showed direct anti-cancer effects against drug-resistant colorectal cancer cells. Rational application of these findings will improve the clinical efficacy of colorectal cancer.
