摘要
以菌丝球数量、菌丝球大小、菌丝体生物量及胞内多糖含量为评价指标,筛选香菇发酵的液体种子培养基和发酵培养基,考察了不同补糖方式对香菇摇瓶补料分批发酵的影响。结果表明,适合香菇发酵的液体种子培养基和发酵培养基(g·L-1)为:葡萄糖10,酵母浸粉20,玉米粉15,磷酸二氢钾0.3,硫酸镁0.15,维生素B10.02。在香菇摇瓶补料分批发酵的第6d和第9d分别补加0.25%葡萄糖,香菇菌丝体生物量达到约847mg·(100mL)-1,胞内多糖含量达到约173mg·(100mL)-1,较不加糖时分别增加了约21.79%和34.29%。补料分批发酵能够提高香菇菌丝体生物量和胞内多糖含量,为香菇多糖大规模工业化生产奠定了基础。
Using mycelium pellet number, mycelium pellet size, mycelium biomass, and intracellular polysac- charide content as evaluation indexes,we screened the liquid seed and fermentation media for the fermentation of Lentinus edodes ,and investigated the effect of different glucose-feeding modes on the fed-batch fermentation of Lentinus edodes in shake flask. Results showed that the optimal liquid seed and fermentation media were as follows(g L-1) :glucose 10,yeast extract 20,corn flour 15,KH2PO4 0.3,MgSO4 0.15,and VB1 0.02. When 0.25 % of glucose was fed at the 6th day and 9th day in the process of fed-batch fermentation of Lentinus edodes in shake flash,the mycelium biomass reached about 847 mg (100 mL)-1 and the intracellular polysaccharide content reached about 173 mg (100 mL)-1 ,which increased by about 21.79% and 34.29% ,respectively,com- pared to those by non-glucose-feeding fermentment. Fed-batch fermentation can improve the mycelium biomass and intracellular polysaceharide content,which lays a foundation for large-scale industrial production of lentin- an.
