摘要
目的:探究不同浓度三氧化二砷(As_2O_3)对肝癌HepG2细胞侵袭转移能力的影响以及对HepG2细胞中USP22蛋白表达的影响,进一步完善其分子机制。方法:CCK8细胞增殖实验检测不同浓度As_2O_3在不同时间点对Hep G2细胞增殖的影响;细胞划痕和Transwell方法检测HepG2细胞在不同浓度As_2O_3作用下侵袭转移能力的变化;Western blot检测As_2O_3作用下HepG2细胞中USP22蛋白的表达水平。结果:随着As_2O_3浓度和作用时间的增加,HepG2细胞增殖明显受到抑制。与对照组相比,24 h时分别用2、4、8μmol/L的As_2O_3处理的HepG2细胞侵袭转移能力降低,细胞中USP22蛋白表达水平明显下降。结论:As_2O_3可抑制人肝癌HepG2细胞侵袭转移能力,其分子机制可能与下调的USP22蛋白水平相关。
Objective To investigate the effects of arsenic trioxide ( As2 O3 ) of various concentrations on invasion, migration and USP22 expression in human HepG2 hepatocarcinoma cells and the underlying molecular mechanism. Methods : CCK8 was used to test the proliferation levels of HepG2 cells treated with As2O3 of different concentrations in different time. Wound - healing and Transwell assays were used to examine the effects of As203 of different concen-trations on cell migration and invasion. The protein levels of USP22 treated with As2 03 of different concentrations were determined by Western blot. Results: With the increase of As20 3 concentration and time,the proliferation of HepG2 cells was obviously inhibited. The migration, invasion and the expression of USP22 protein in HepG2 cells which were treated with 2,4,8 jjimol/L As2O3 respectively for 24 h were obviously decreased compared with the control group. Conclusion : As2 O 3 could inhibit the migration and invasion of human hepatocellular carcinoma HepG2 cells, which may be related to the down - regulation of USP22 protein.
