摘要
目的:探讨助孕Ⅰ号方孕前直肠给药对肾虚血瘀模型大鼠子宫蜕膜化及其组织层粘连蛋白(LN)、integrinβ3表达的影响。方法:选取动情前期的健康未交配雌性SD大鼠60只,随机分为空白组、模型组、助孕Ⅰ号方低剂量组、助孕Ⅰ号方高剂量组、五子组和丹参组。妊娠第8天处死大鼠,剖取子宫并制片,采用免疫组化、HE染色。结果:与空白组比较,模型组的大鼠妊娠第8天子宫蜕膜组织LN面积总和、子宫蜕膜组织大部分蜕膜细胞在形态、排列、细胞核、血管等方面的表现呈劣势,差异有统计学意义(P<0.01)。与模型组比较,助孕Ⅰ号方低剂量组、助孕Ⅰ号方高剂量组子宫蜕膜组织细胞在形态、排列、细胞核等方面的状况呈优势,并且助孕Ⅰ号方低剂量组大鼠妊娠第8天子宫蜕膜组织LN的面积总和及助孕Ⅰ号方高剂量组子宫蜕膜组织LN面积总和、integrinβ3面积总和明显增加,差异有统计学意义(P<0.05,P<0.01)。结论:助孕Ⅰ号方(即补肾活血法中药)孕前直肠给药可改善肾虚血瘀模型大鼠子宫蜕膜组织形态,提高子宫蜕膜组织LN、integrinβ3的表达,从而有利于胚泡着床,增加妊娠率。
Objective：To explore the effect of rectal administrationof enema-assisted pregnancy Ⅰ prescription before pregnancy on uterine decidualization and the expression of LN and integrinβ3 in kidneydeficiency and bloodstasis model rats.Methods：A total of 60 pre-estrous female SD virgin rats were randomly divided into blank group,model group,assisted pregnancy Ⅰ prescription low dose group,assisted pregnancy Ⅰ prescription high dose group,Wuzi group and Salvia miltiorrhiza group.The rats were sacrificed on the 8 th day of pregnancy.The uterus was cut and made into slices.Immunohistochemistry and HE staining were performed.Results：Compared with the blank group,on 8 th day pregnancy the model group rats＇ uterine decidua LN area sum were decreased,and most decidual cells in the uterine decidua in the model group showed inferiorities in morphology,arrangement,nucleus,blood vessel and so on,and the difference was statistically significant（P〈0.01）.Compared with the model group,uterine decidual tissue cells of assisted pregnancy Ⅰ prescription lowdose group and assisted pregnancy Ⅰ prescription highdose group showed superiorities in morphology,arrangement,the nucleus and other aspects.And the uterine decidua LN area sum of assisted pregnancy Ⅰ prescription lowdosegroup,the uterine decidua LN area sum and integrinβ3 area sum of pregnancy Ⅰprescription highdose group increased significantly,and the difference was statistically significant（P〈0.05,P〈0.01）.Conclusion：Assisted pregnancy Ⅰ prescription（tonifying kidney and activating blood therapy） rectal administration pre-pregnancy can improve the morphology of endometrial decidualization and angiogenesis of kidney deficiency and blood stasis model rats.It can improve the expressions of LN and integrinβ3 in the decidua of uterus,which is favorable for implantation of blastocyst and increases the pregnancy rate.
