表皮生长因子受体对脂多糖诱导大鼠急性肺损伤的抑制作用及对炎症反应的影响 被引量:9

摘要 目的探讨表皮生长因子受体在脂多糖诱导大鼠急性肺损伤的抑制作用及对炎症反应的影响。方法选择2014年6月至2016年7月进行实验的SD大鼠40只,取10只大鼠作为空白对照组,另取30只大鼠采用脂多糖诱导建立大鼠急性损伤模型。建模成功后随机数字法分为阳性对照组(n=10)、生理盐水组(n=10)和表皮生长因子受体组(n=10)。阳性对照组建模成功后不采取任何措施处理,生理盐水组在大鼠建模成功后注射0.5 ml/kg生理盐水,表皮生长因子受体组建模成功后注射0.5 mg/kg表皮生长因子受体抑制剂。采用酶联免疫吸附试验测定各组大鼠肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平;取大鼠急性肺损伤组织进行病理HE染色,测定组织中过氧化物酶(MPO)含量、肺湿/干重比(W/D)。结果表皮生长因子受体组与空白对照组建模处理3 d后TNF-α及IL-6水平差异无统计学意义(P>0.05);表皮生长因子受体组TNF-α及IL-6水平均低于生理盐水组和阳性对照组(P<0.05);生理盐水组TNF-α及IL-6水平,均低于阳性对照组(P<0.05)。空白对照组未参与建模,肺部未见损伤,HE染色下正常;阳性对照组建模成功后未采取措施处理,肺部损伤明显,炎症反应严重;生理盐水组肺组织中损伤明显,存在肺气肿、肺泡塌陷等典型组织变化;表皮生长因子受体组大鼠肺部组织病理变化明显改善;表皮生长因子受体组MPO及W/D水平,均低于生理盐水组、阳性对照组(P<0.05);生理盐水组MPO及W/D水平,均低于阳性对照组(P<0.05)。结论脂多糖能诱导大鼠急性肺损伤,将表皮生长因子受体抑制剂用于大鼠急性肺损伤中有助于降低炎症反应,值得推广应用。 Objective To discuss the inhibition effect of epidermal growth factor receptor(EGFR) and its impact in the inflammatory response for rats with lipopolysaccharide-induced acute lung injury.Methods 40 SD rats tested from June 2014 to July 2016 in our hospital were selected.10 rats were selected as control group;30 rats were selected to establish the models of lipopolysaccharide-induced acute lung injury.After modeling made,the subjects were randomly divided into positive group(10 cases),NS group(10 cases) and EGFR group(10 cases).The positive group didn't take any measures.The NS group took the 0.5 ml/kg of normal saline;the EGFR group took the 0.5 mg/kg of EGFR antagonists.The index of TNF-α and IL-6 were detected by enzyme linked immunosorbent assay.The index of MPO and W/D were detected by pathological HE staining.Results After 3 d of modeling,the index of TNF-α and IL-6 of EGFR group and control group were not significantly different(P > 0.05).The index of TNF-α and IL-6 of EGFR group were lower than those of NS group and positive group(P < 0.05).The index of TNF-α and IL-6 of NS group were lower than those of positive group(P < 0.05).Since the model was not established,the control group had no lung injury and abnormal HE staining;without the proper measures,the positive group had the severe lung injury and inflammatory response.The NS group had the severe lung injury with the classical pathological changes of pulmonary emphysema and alveolar collapse;after the treatment of EGFR,the pathological changes were improved.The index of MPO and W/D of EGFR group were lower than NS group and positive group(P < 0.05).The MPO and W/D for NS group were lower than positive group(P < 0.05).Conclusion For rats with lipopolysaccharide-induced acute lung injury,the EGFR antagonists are good to reduce the inflammatory response.It is worthy of clinical application.
