摘要
采用RT-PCR法,检测了万载龙牙百合的主要病毒;通过RT-PCR反应条件的确定,建立了万载龙牙百合主要病毒的RT-PCR检测技术。结果表明:万载龙牙百合感染了百合斑驳病毒,没有感染黄瓜花叶病毒和百合无症病毒;百合斑驳病毒的RT-PCR检测技术为:50℃逆转录30 min;94℃预变性5 min;94℃变性30 s,60℃退火30 s,72℃延伸30 s,30个循环;72℃最后延伸7 min。
The main viruses of Lilium brownii var. viridulum in Wanzai were detected by using RT-PCR method,and their detection technology was established by optimizing the conditions for RT-PCR. The results indicated that L. brownii var. viridulum in Wanzai was infected by lily mottle virus,but was not infected by cucumber mosaic virus and lily symptomless virus. The RTPCR detection technology for lily mottle virus was established as follows: reverse transcription for 30 min at 50 ℃; pre-denaturation for 5 min at 94 ℃; denaturation for 30 s at 94 ℃,annealing for 30 s at 60 ℃,extension for 30 s at 72 ℃,30 cycles; final extension for 7 min at 72 ℃.
