摘要
以盆栽、大棚以及石头仿野生、仿野生贴树栽培等4种栽培方式的铁皮石斛为研究对象,采用常规方法测试生物学形态特征与多糖积累情况,HPLC方法检测葡萄糖(Glc)、甘露糖(Man)、半乳糖(Gal)、木糖(Xyl)等4个单糖含量。结果为4种栽培方式中收获枝条生物量为大棚>岩石仿野生>仿野生贴树干>盆栽;仿野生贴树干栽培多糖含量最高,达到31.6540%,尤其是半乳糖含量较高,占总糖含量的79.73%;岩石仿野生栽培方式的含水量最低,为71.69%,其甘露糖含量最高,为0.00961mg/ml;TOPSIS法综合评价4种栽培模式优劣依次为:岩石仿野生>大棚>仿野生贴树干>盆栽,仿野生两种模式优于其它两种设施栽培。鉴于该地区两种林下仿野生栽培模式中岩石栽培所产的铁皮石斛品质相对较优,可作为该地区大面积推广的较佳栽培方式。
To screen out the cultivation mode of high quality product,we adopt comparison of the contents of polysaccharide and monosaccharide composition of Dendrobium officinale under different cultivation methods.An HPLC method was established for simultaneous determination of four monosacchride compounds(Glucose,Mannose,Galactose,Xylose) to investigat the planting pattern of flowerpot(Ⅰ),the pattern of greenhouse(Ⅱ),the wild living on rock(Ⅲ) and the wild living on tree stem(Ⅳ).The biomass were the pattern of greenhouse > the wild living on tree stem > the wild living on rock > lowerpot.The main characteristics of the wild living on tree stem were the highest content of polysaccharide,reaching 31.6540%,especially the high content of galactose,accounting for about 79.73% of the total sugar content.The wild living on rock cultivation method had the lowest moisture content(71.69%) and the highest content of mannose(0.00961 mg/ml).The order of TOPSIS evaluation to 4 plan-ting patterns was Ⅲ > Ⅱ > Ⅳ > Ⅰ.method.The best quality of product is the model cultivation of the wild living on rock and on tree stem,which can be used as a better method of cultivation in the area.
