摘要
[目的]构建脊柱结核小鼠模型并分析其椎体的骨密度改变。[方法]60只8周龄雌性C57BL/6小鼠随机分成实验组和对照组,每组30只。实验组小鼠脊柱局部注射浓度为108CFU/ml的H37Rv标准菌株悬液0.1 ml,对照组注射生理盐水0.1 ml。饲养12周后取脊柱标本送Micro-CT扫描。获得三维松质骨图像后,评估两组小鼠椎体骨小梁数量及骨小梁厚度差异。[结果]共6只小鼠在Micro-CT上发现了脊柱破坏的病灶。破坏的椎体骨小梁数及骨小梁厚度显著低于未受累的椎体(t=-5.710,P<0.001)和对照组相同节段椎体(t=-6.771,P<0.001),脊柱结核小鼠的椎体平均骨小梁数量较对照明显下降(t=-4.716,P<0.001)。虽然病灶椎的总体骨小梁数量及厚度降低,但在病灶周围存在大量硬化骨形成,病灶局部骨小梁数量及厚度显著增高(t=8.987,P<0.001)。[结论]小鼠脊柱结核病椎平均骨小梁密度下降,但病灶局部骨小梁密度增高。
[Objective] To build a spinal tuberculosis model in mice and analyze the changes of vertebral bone mineral density. [Methods] Sixty 8-weeks C57BL/6 mice were randomly divided into experimental group and control group with 30 animals in each group. As treatment, 0.1 ml suspension of the H37Rv standard tuberculosis strain in 108CFU/ml was injected into the spine of the experimental group, whereas 0.1ml normal saline given in the control group. After 12 weeks, the specimens of spine were examined by Micro-CT. The difference of trabecular number and trabecular thickness between the two groups were evaluated via three-dimensional imaging. [Results] Destructive lesions were note in 6 mice on the micro-CT images. The trabecular number and trabecular thickness in the affected vertebra were significantly lower than the adjacent unin- volved vertebra (t=-5.71, P 〈 0.001) , likewise, considerably lower than the same segment vertebra in the control group (t=- 6.771, P 〈 0.001). In addition, tile average number of trabecul^r bone in vertebral body of the mice with tuberculosis was signifi- candy lower than the control group (t=-4.716, P 〈 0.001). Although the trabecular number and trabecular thickness were re- duced in the lesion, there was a large amount of sclerotic bone with significantly increased trabecular number and trabecular thickness around the lesion (t=8.987, P〈0.001) . [Conclusions] The density of average trabecular bone was decrease in the pathologic vertebra, but the density of local trabecular bone was increase.
