耐盐碱刺槐离体再生快繁体系的建立与优化 被引量:2

Establishment and optimization of rapid regeneration system in vitro for salt-tolerance Robinia pseudoacacia

摘要 为提高耐盐碱刺槐的快繁效率,满足沿海及滨海盐碱地区绿化要求,以耐盐碱刺槐无性系"W009"和"W038"为试材,研究不同条件对耐盐碱刺槐试管苗分化和生根的影响.结果表明:两刺槐茎尖初代接种不定芽萌发率均好于茎段,"W009"离体茎尖不定芽萌发的最适培养基为MS+6-BA 0.5 mg·L^(-1)+NAA 0.05mg·L^(-1),茎段不定芽萌发的最适培养基为MS+6-BA 1.0mg·L^(-1)+NAA 0.1mg·L^(-1);而适合"W038"离体茎尖和茎段不定芽萌发的最适培养基则为MS+6-BA 1.0mg·L^(-1)+NAA 0.1 mg·L^(-1);适合"W009"不定芽增殖的培养基为MS+6-BA 1.0mg·L^(-1)+NAA 0.05mg·L^(-1),"W038"不定芽增殖的培养基为MS+6-BA 2.0mg·L^(-1)+NAA 0.1 mg·L^(-1);"W009"最适生根培养基为:1/2 MS+0.1 mg·L^(-1) IBA+0.2mg·L^(-1) NAA+1.0g·L^(-1)活性炭;"W038"最适生根培养基为:1/2 MS+0.3 mg·L^(-1) IBA+0.2 mg·L^(-1) NAA+1.0g·L^(-1)活性炭. To improve the efficiency of rapid propagation of salt-tolerance Robinia pseudoacacia and meet requirements of green in coastal and coastal saline area, the effects of the different conditions on plantlet differentiation and rooting were studied using salt- tolerance black locust- "W009" and "W038" as the experimental materials. The results showed that the germination rate of adventitious buds for shoot tip was better than that of stem segments, and the optimum medium of "W009" for adventitious bud germination from shoot tip was MS + 6-BA 0.5 mg · L^-1+ NAA 0.05 mg · L^-1, the medium MS+6-BA 1.0 mg · L^-1+NAA 0.1 mg · L^-1 was the best for stem segments; while the optimal medium for germination of adventitious buds from shoot tips and stem segments of "W038" was MS + 6-BA 1.0 mg · L^-1 NAA 0.1 mg · L^-1. The culture medium of "W009" for adventitious bud proliferation was MS q- 6-BA 1.0mg · L^-1+ NAA 0.05 mg · L^-1 , and themedium MS + 6-BA 2.0 mg · L^-1+- NAA 0.1 mg · L^-1 was optimal for "W038". The rooting medium 1/2 MS medium containing 0.1mg · L^-1 IBA, 0.2 mg · L^-1 NAA and 1.0 g · L-1 AC was the most suitable for "W009", and the optimum rooting medium of "W038" was 1/2 MS medium combined with 0.3 mg · L^-1 IBA, 0.2 mg · L^-1 NAA and 1.0 g·L-1AC.
