辛伐他汀对高胆固醇血症小鼠血管内皮粘附单核细胞的抑制作用 被引量:9

摘要 目的 :观察辛伐他汀对高胆固醇血症小鼠离体胸主动脉内皮粘附单核细胞的抑制效应。方法 :用不同剂量 (1,5 ,2 0mg·kg- 1)的辛伐他汀每日1次给高胆固醇血症小鼠皮下注射共 2wk或 6wk ,届时剪取小鼠胸主动脉 ,用于单核细胞粘附试验。结果 :(1)只有 2 0mg·kg- 1× 6wk组小鼠血浆胆固醇水平显著降低 ,从 6 .6± 0 .7mmol·L- 1降为 4 .6± 0 .7mmol·L- 1,(P <0 .0 5 ) ;(2 )高胆固醇血症小鼠胸主动脉内皮对单核细胞的粘附率明显高于正常小鼠 ,辛伐他汀 2 0mg·kg- 12个组对高胆固醇血症小鼠胸主动脉内皮粘附单核细胞有明显的抑制作用 ,粘附率 (4.1± 2 .5 ) %vs (1.9± 0 .1) % (和 )或(1.4± 0 .3) %均 (P <0 .0 5 )。结论 :高胆固醇血症小鼠胸主动脉内皮对单核细胞粘附增强 。 AIM: To observe the inhibitory effect of simvastatin on the vascular endothelial adhesiveness for monocyte in hypercholesterolemic mice. METHODS: Mice with apolipoprotein E-knockout were treated with different dose(1,5,20 mg·kg -1) of simvastatin subcutaneously for 2 wk or 6 wk. Thoracic aortae were harvested and endothelial adhesiveness for monocyte was assessed by functional binding assay. RESULTS: Plasma total cholesterol level in 20 mg·kg -1×6 wk group decreased after 6 wk of treatment (P<0.05). There was an increased monocyte adherence to the endothelium in hypercholesterolemic mice, and simvastatin 20 mg·kg -1 inhibited the enhanced endothelial adhesiveness for monocyte in hypercholesterolemic mice significantly(P<0.05). CONCLUSION: There is an enhanced endothelial adhesiveness for monocyte in apolipoprotein E-knockout mice and simvastatin may improve the impaired endothelial adhesive function. The inhibitory effect of simvastatin could not be attributed to its lipid-lowering effect.
