一种鉴定常见分枝杆菌分离株的简单快速分子生物学方法

摘要 目的 :用分子生物学方法检测常见的分枝杆菌临床分离株 ,以适应临床分枝杆菌标本鉴定的需要。方法 :对 11株分枝杆菌参考株及 15 0株临床分离株用PCR RFLP的方法分析其hsp6 5基因。结果 :对结核分枝杆菌复合群和鸟分枝杆菌复合物鉴定的敏感性和特异性达 10 0 %。结论 :该方法简单、快速、经济 ,特别适用于标本量较大的临床实验室对分枝杆菌的鉴定。 Objective To develop a novel PCR restriction fragment length polymorphism analysis (PRA) of the hap65 gene. Methods One hundred and sixty one specimens (11 reference striains and 150 clinical isolates) were tested, of which 80 were Mycobacterium Tuberculosis Complex (MTC) and 39 were Mycobacterium Avium Complex (MAC). Results The restriction patterns for MTC and MAC were designed to be highly distinct and the overall number of restriction patterns was limited. The assay was virtually 100%sensitive and specific for MTC and MAC. Conclusion This assay could be completed on the same day, and is user friendly and rapid. Therefore, it is highly suitable for the large scale use in a clinical laboratory.
