摘要
目的探讨hsp65基因序列PCR-RFLP法用于临床分枝杆菌鉴定的诊断价值。方法用hsp65基因序列PCR-RFLP法鉴定经过传统方法鉴定过的130株临床分枝杆菌。结果130株临床分枝杆菌中,鉴定出结核分枝杆菌105株,戈登分枝杆菌4株、瘰痢分枝杆菌3株、耻垢分枝杆菌1株。用PCR-RFLP法,偶然分枝杆菌复合群和鸟胞分枝杆菌复合群进一步鉴定,偶然分枝杆菌2株、脓肿分枝杆菌2株、龟分枝杆菌4株、鸟分枝杆菌5株、胞分枝杆菌4株。结论hsp65基因序列PCR-RFLP法比传统方法有更高的准确性、敏感性、特异性和重复性,具有很好的临床应用价值。
Objective To explore the diagnostic value of PCR-RFLP analysis of the hsp 65 gene. Methods PCR-RFLP analysis of the hsp 65 gene was applied to 130 mycobacterial isolates that were identified by the traditional methods. Results Of 130 Mycobacterial Isolates, we obtained M. tuberculosis ( n=105 ), M. gordonae ( n = 4 ) , M. scrofulaceum ( n = 3 ) , M. smegmatis ( n = 1 ), and M. avium -M. intracellulare complex and M. fortuitum complex were discriminated to the spccles level, M. avium ( n = 5 ) , M. intracellulare (n=4) , M. fortuitum (n=2), M. chelonei (n=4) , M, abscessus (n=2). Conlusions PCR-RFLP analysis of the hsp65 gene has good clinical application. Its sensitivity, specificity, accuracy and repetition are more than the ones of the traditional methods.
