急性高原低压缺氧对成年大鼠海马齿状回细胞增殖和分化的影响 被引量:1

摘要 目的模拟高原低压缺氧环境,观察急性低压缺氧刺激对大鼠海马齿状回颗粒下区神经干细胞增殖和分化的影响。方法 120只健康成年SD大鼠,随机分成两组:模拟海拔5 000米组(实验组)和海拔2 200米组(对照组);实验组在模拟海拔5 000米低压氧舱内预处理3天,进行急性高原低氧损伤造模,然后转移至海拔2 200米环境下行常规饲养,对照组不经低压氧舱预处理,直接在海拔2 200米环境下行常规饲养。实验组和对照组在2 200米环境下常规饲养1、3、7、14、28天后进行组织取材,分别用免疫组织化学和免疫荧光染色,显微镜下观察BrdU、BrdU/DCX、BrdU/GFAP阳性细胞表达并行相关分析。结果模拟急性高原低压缺氧刺激后3、7天组齿状回的BrdU阳性细胞显著增多(P<0.001),14天组BrdU阳性细胞依然明显高于对照组(P<0.01)。急性高原低压缺氧刺激后1、3、7天组DCX阳性细胞表达增加,其中3天组DCX阳性细胞表达显著增加(P<0.001);但BrdU/DCX双阳性细胞比例和对照组没有差别。急性高原低压缺氧刺激后1、3天组GFAP阳性细胞数显著增加,多于对照组(P<0.001),在缺氧刺激后BrdU/GFAP双阳性细胞率高于对照组[1天组(P<0.05)、3天组(P<0.01)、7天组(P<0.05)]。结论急性高原低压缺氧可促进成年大鼠海马齿状回神经干细胞增殖、维持神经前体细胞向神经元高分化状态,并促进海马齿状回星形胶质细胞活化。 Objective To investigate the proliferation and differentiation of Subgranular zone (SGZ) cells in the hippoeampus dentate gyms at high altitude hypoxia stimulation in adults rat.Methods 120 healthy adult SD rats were randomly divided into two group : simulated altitude of 5 000 meters group ( experimental group) and 2 200 me- ters group(control group).Experimental group was pretreatedwith low pressure oxygen tank for 3 days to simulate the 5 000 meters environment. After that the animals were transferred to an altitude of 2 200 meters environment. Control group was lived directly in an altitude of 2 200 meters environment.Two group divided into five subgroup af- ter acute hypobaric hypoxia: 1 d,3 d,7 d, 14 d, and 28 d, respectively.Immunohistochemical staining and immuno- fluorescence staining to detect the BrdU, BrdU/DCX, BrdU/GFAP positive cells in SGZ of hippocampal dentate gy- ms and positive cells were counted.Results The BrdU positive ceils of experimental group were increased than that of the control group in 3 days and 7 days after acute hypobaric hypoxia, the number of BrdU positive cells in SGZ increased significantly compared with the control group( P〈0.001 ) , at the day of 14 after acute hypobaric hypoxia, the BrdU positive cells were still higher than that of control group(P〈0.01 ).The DCX positive cells increased at 1 d ,3 d and 7 d after acute hypobaric hypoxia, and the DCX positive cells increased significantly in 3 d than control group( P〈0.001 ) ,but there was no difference of BrdU/DCX double positive cells ratio between experimental group and control group.The number of GFAP positive cells at 1 d and 3 d after acute hypoxia was significantly higher than that control group (P〈0.001).After hypoxia stimulation, the rate of BrdU/GFAP double positive cells was high- er than control group 1 d ( P〈0.05), 3 d ( P 〈 0.01 ), and 7 d ( P 〈 0.05 ). Conclusion Acute hypobaric hypoxia can promote the proliferation of SGZ neural stem cells in hippocampus of adult rats, and promote the differentiation of SGZ precursor cells into neurons.Acute hypobaric hypoxia promote the activation of astrocytes in dentate gyms.
