摘要
本文以牛血红蛋白为底物,在一定条件下进行酶促反应、用酪氨酸作对照,紫外分光光度法测定胃蛋白酶合剂中的酶活力。该法简便、快速,精密度好,CV2.3%(n=9),适合医院制剂的快速分析。
A rapid and simple assay method for the de-termination of enzyme actity in pepsin mixture, ahospital dispensary preparation, involyed the enz-matic reaction using hemoglobin (bovine blood) asa substrate. After incubating for 10 minutes at 37±0.1℃ in hydrochloric acid solution,trichloroacetic acid solution was added. The solutionwas mixed, filtered and determined by UV absorp-tion spectrophotometry at 275±1 nm. One unit willproduced 1.00μmol of tyrosine from hemoglobinper unit at pH 2.0 at 37℃, CV 2.3% (n=9).
