摘要 目的 采用骨髓间质干细胞 (MSCs)体外转化为肌细胞 ,为心衰的干细胞移植治疗提供新的细胞材料。方法 抽取贵州香猪骨髓液 3ml,按照Wakitani的方法培养骨髓间质干细胞 ,经5 氮胞苷 (5 azacytidine)刺激 2 4h后 ,进行结蛋白 (desmin) ,肌球蛋白重链 (MHC)免疫组织化学 ,透射电镜鉴定。结果 骨髓间质干细胞经 5 aza刺激后 ,部分细胞呈梭形 ,结蛋白 ,肌球蛋白重链阳性 ,有多核肌管形成 ,透射电镜可见明显的肌丝形成。结论 骨髓间质干细胞可在体外的环境下向肌细胞转化 。 Objective By using myogenic cells from porcine marrow mesenchymal stem cells (MSCs), to establish a new cell source of stem cell transplantation for heart failure therapy. Methods 3 ml bone marrow was extracted from the posterior superior iliac spine of Gui zhou Xiang porcine. Mesenchymal stem cells were cultured according to the methods of Wakitani's. After being co cultured with 5 azacytidine for 24 h, the cultured cells were evaluated by a transmission electron microscope and immunohistochemically stained for myosin heavy chain (MHC) and desmin.Results After being co cultured with 5 azacytidine, some of MSCs became spindle like and formed a network of myotubules. Myotubular cells were found to be stained with positively MHC and desmin. Transmission electron microscopy showed that myofilaments were formed. Conclusion Myogenic cells can be generated from MSCs in vitro, suggesting that MSCs may be a new cell source for stem cells transplantation in heart failure therapy.
