摘要 目的探讨MSCs共培养抗ARPE-19细胞氧化应激损伤的作用。方法常规培养人RPE细胞系ARPE-19,分为正常组、H_2O_2组和共培养组;通过观察细胞形态变化、检测细胞ROS水平和调亡水平研究MSCs共培养的抗ARPE-19细胞氧化应激损伤作用。结果 H_2O_2诱导的氧化应激损伤能影响细胞形态,引起细胞内ROS与凋亡水平显著增高;而MSCs共培养预能减轻H_2O_2诱发的细胞形态变化,并逆转ROS与凋亡水平增高,以上差异均具有统计学意义(P<0.05)。结论 MSCs共培养能对抗H_2O_2诱导的人视网膜上皮细胞损伤发挥保护作用。 Objective To investigate the protective effect of co-culture of MSCs on oxidative stress injury in ARPE-19 cells.Methods The ARPE-19 cell was routinely cultured and divided into normal group,H2O2 group and co culture group. we studied the effect of co-culture of MSCs on the oxidative stress injury by observing the morphological changes of cells,detecting the level of ROS and the level of apoptosis. Results The oxidative stress injury induced by H2O2 can affect the morphology of cells,and increase the level of ROS and apoptosis in cells. However,co-culture of MSCs can reduce the morphological changes and reverse the increase of ROS and apoptosis level induced by H2O2(P〈0.05). Conclusion The co-culture of MSCs can protect human retinal epithelial cells from injury induced by H2O2.
