摘要
以汪清县兰家林场采集的笃斯越桔叶片为样本,采用PCR方法扩增rbcL基因,分析rbcL序列在笃斯越桔及其近缘植物的序列差异,分析其系统发育关系。结果表明:克隆得到的笃斯越桔rbcL基因片段长599bp,编码199个氨基酸残基。核苷酸多序列比对发现,笃斯越桔与欧洲越桔和小叶越桔在第68和390碱基位置有差异,并从构建的系统发育树中得出笃斯越桔和卵叶越桔首先聚为一类,说明笃斯越桔和卵叶越桔亲缘关系更近。
Leaves of the Vaccinium uliginosum collected from Wangqing Lanjia forest as materials,the rbcLgene was cloned by PCR,and its sequence and phylogenetic relationships in comparison with rbcLsequences from related Vaccinium plants were analyzed.The results,showed that the length of DNA fragment of rbcLgene fromVaccinium uliginosum was 599 bp,encoding apeptide fragment comprising of 199 amino acids.The nucleotide sequence isolated here involved two single base differences at the 68 th and390 th bases compared the DNA sequences of rbcLgene fromVaccinium myrtillus and Vaccinium parvifolium,respectively.By DNA sequence alignment,Vaccinium uliginosum and Vaccinium ovatumwas clustered into the same class in the phylogenetic tree,indicating that the Vaccinium uliginosum was more closely related with Vaccinium ovatum.
