摘要
为了分析fat-1转基因小鼠对3个脂类代谢关键基因(FASN、PPAR、LPL)的组织表达特性,研究利用实时荧光定量PCR技术检测FASN、PPAR和LPL在心脏、肝脏、骨骼肌组织中的表达情况。结果表明:FASN、PPAR、LPL基因在转基因小鼠心脏、肝脏、骨骼肌的表达情况有很大差异。其中PPAR和LPL基因在转基因小鼠肝脏组织中极显著升高(P<0.01),而在心脏、骨骼肌的表达量显著或极显著下降(P<0.01或P<0.05);FASN基因只在骨骼肌中极显著下降(P<0.01),而在心脏和肝脏中没有发生明显变化(P>0.05)。说明fat-1基因能够在机体脂类代谢的调控过程中发挥重要的作用。
The aim of the present study was to analyze the tissue expression characteristics of three key lipid metabolism genes (FASN, PPAR and LPL) in the fat - 1 transgenic mice. The expression of FASN, PPAR and LPL in heart, liver and skeletal muscle tissue were detected by real - time PCR, respectively. The results showed that the expression differenee of FASN, PPAR and LPL gene were significantly different be- tween transgenic mouse heart, liver and skeletal muscle. The expression of PPAR and LPL genes were significantly increased in the liver tissues of transgenic mice ( P 〈 0.01 ), while the expression of PPAR and LPL in the heart and skeletal muscle was significantly decreased ( P 〈 0.01 or P 〈 0.05 ). The expression of FASN gene was significantly decreased only in skeletal muscle (P 〈 0.01 ), but not in the heart and liver (P 〉0.05 ). The results indicated that the fat - 1 gene could play an important role in the regulation of lipid metabolism in the body.
