补泻不同配伍补阴方对衰老大鼠抗氧化能力及Bax、Bcl-2、Caspase-3蛋白表达的影响 被引量:2

摘要 目的比较左归丸和六味地黄丸及共有"三补"药物组抗衰老的作用机制。方法观察左归丸和六味地黄丸及共有"三补"药物组对D-半乳糖(D-gal)致亚急性衰老大鼠血清和肾组织的超氧化物歧化酶(SOD)、丙二醛(MDA)含量及对肾组织Bax、Bcl-2、Caspase-3蛋白表达的影响。结果亚急性衰老大鼠SOD水平下降,MDA含量升高,Bax、Bcl-2、Caspase-3蛋白表达水平上调;对于血清中的SOD活性,左归丸和六味地黄丸均有显著的升高作用,"三补"的升高作用显著;对于肾组织中的SOD活性,各给药组均无升高作用;对于血清和肾组织中的MDA,三给药组均有明显的降低作用;两补肾方和"三补"药物组均有下调肾组织促凋亡因子Bax、Caspase-3蛋白表达和上调抗凋亡因子Bcl-2蛋白表达的作用。结论两补肾方可能是通过提高SOD活力、降低MDA含量和降低Bax、Caspase-3以及升高Bcl-2表达而达到延缓衰老的效果。左归丸和六味地黄丸尽管存在补泻配伍方式的不同,但在抗衰作用上差异不显著;补泻兼顾和纯补无泻的组方思想只是在具体的药理机制上有所不同。 Objective To analyze and compare the anti-aging function's mechanisms of the two invigorating yin formulas,Zuogui Pill and Liuweidihuang Pill.Methods The expressions of superoxide dismutase( SOD) and malondialdehyde( MDA) in serum and kidney tissue,and the expressions of Bax,Bcl-2 and Caspase-3 protein in the kidney tissue of subacute aging rats induced by D-galactose were observed.Results The level of SOD was reduced,the levels of MDA,the expressions of Bax,Bcl-2 and Caspase-3 were increased in subacute aging rats. The activity of SOD in serum was stepped up significantly by Zuogui Pill,Liuweidihuang Pill,also,it was rised obviously by Sanbu group. But,there was no significant difference on the activity of SOD in kidney tissue among three groups. Wherever,the MDA in serum and kidney tissue was cut down significantly in three groups. The expressions of Bax and Caspase-3,pro-apoptotic factors were cut down and the expressions of Bcl-2,anti-apoptotic factors,were stepped up in the two tonifying kidney formulas and Sanbu group. Conclusions The thought of the combination of supplementation and draining and the pure supplement are only different in the specific pharmacological mechanism.
