摘要
目的:比较龟鹿二仙胶及其拆方含药血清对硝普钠(SNP)诱导后软骨细胞bax、bcl-2表达的影响,探讨龟鹿二仙胶的配伍机理。方法:取第三代4周龄SD大鼠关节软骨细胞,经SNP诱导凋亡后,采用免疫组化染色法及Realtime-PCR观察龟鹿二仙胶及其拆方含药血清对软骨细胞bax、bcl-2表达的影响。结果:龟鹿二仙胶及其拆方各组均可引起bcl-2表达升高及bax下降,与生理盐水组比较差异有统计学意义(P<0.05),龟鹿二仙胶与龟板、鹿角比较差异无统计学意义(P>0.05)。结论:龟板、鹿角、人参、枸杞均具有抑制软骨细胞凋亡的作用,而4味药同用,人参、枸杞并无显著加强龟板、鹿角的抑制凋亡作用。
Objective:To investigate the mechanism of GuiLu ErXian Glue in Rats'chondrocyte apoptosis induced by nitroprussid,by detecting the expression of bax/bcl2in rats'chondrocyte induced by nitroprussid treated with Guiluerxian glue and its serum reconstituent.Methods:Chondrocyte were extracted from rat's knee and cultured in vitro.Apoptosis of chondrocyte were induced by nitroprusside.The expressions of bax/bcl2were detected by immunohistochemistry stain and realtime-PCR.Results:The expression of Bcl-2was increased and the expression of Bax were decreased in the GuiLu ErXian group,there were significant difference in the expression of Bcl-2/Bax between the GuiLu ErXian group and the control group(P0.05).There were no difference in the expression of Bcl-2/Bax between the GuiLu ErXian and the Guiban,Lujiao(P0.05).Conclusions:Tortoiseshell,antler,ginseng and medlar can inhibit the apoptosis of chondrocyte in rat.Inhibition of apoptosis aroused by Tortoiseshell,antler was not strengthened by ginseng and medlar.
