正常人及系统性红斑狼疮患者T细胞亚群流式细胞术初步分析 被引量:5

Flow Cytometric Analysis of Peripheral T Cells from Normal and Systemic Lupus Erythematosus

摘要 目的:研究正常人及系统性红斑狼疮(SLE)患者外周血(PBMCs)T淋巴细胞亚群的平衡状态。方法:采用微量全血法在样本中标记多种荧光抗体,利用三色流式细胞技术对正常人及SLE患者外周血各T细胞亚群百分率进行检测和分析。结果:与正常人相比,在未经刺激的自然状态下,SLE患者PBMCs中表达白细胞介素(IL)-10的Th2细胞及Tc2细胞的百分率均显著增高;如经植物血凝素(PHA)和佛波醇乙酯(PMA)联合刺激,正常人表达干扰素-γ(IFN-γ)的Tc1细胞百分率显著升高,而SLE患者则仍表现为Th2和Tc2细胞百分率的明显增高。结论:SLE患者体内存在高IL-10应答、低IFN-γ应答状态,从而形成T细胞亚群平衡向Th2/Tc2偏移的态势。 Objective:To investigate the imbalance of peripheral T cells subtypes in normals and patients with systemic lupus erythematosus.Methods:Three color flow cytometric analysis with PE Cy5 anti CD4/PE Cy5 anti CD8,PE anti IL 10 and FITC anti IFN γ was employed to measure peripheral T cells in SLE and normal donors with trace blood samples.Results:Unstimulated in vitro,the percentages of Th2 and Tc2 cells expressing interleukin-10 from PBMCs in SLE which were not stimulated with any phytohemagglutinin,are significantly higher when compared with controls.Stimulated with PHA and PMA,the percentage of Tc1expressing IFN γ in the controls was highly increased but the increased level was the same as the unstimulated in SLE.Conclusion:The behavior of high expression of IL-10 and low expression of IFN γ in Th/Tc cells of SLE forms the deriation of Th2/Tc2.
