摘要
目的分析2011年-2016年唐山地区B型流感血凝素基因特征。方法对唐山市哨点医院流感样病例咽拭子标本进行核酸检测,阳性标本采用MDCK细胞分离培养,选取10株B型流感病毒进行HA1基因测序,分析其核苷酸序列特征及氨基酸变异情况。结果进化分析显示,唐山地区B型流感病毒分为B/Victoria系和B/Yamagata系,其中4株B/Vic系毒株为1B分支(2011年-2012年)和1A分支(2015年-2016年),6株B/Yam系毒株均属于3分支。与疫苗株B/Brisbane/60/2008相比,B/Vic系毒株氨基酸替换发生在N129S和I146V(1B分支)、I117V和N129D(1A分支)。与疫苗株B/Wisconsin/1/2010(B/Yam 3分支)相比,B/Yam毒株共有7个位点发生氨基酸替换;与疫苗株B/Massachusetts/2/2012(B/Yam 2分支)相比,有14个位点发生氨基酸替换,其中N116K、S150I、N165Y和D196N位于抗原表位上。结论 2011年-2016年唐山地区B型流感毒株发生明显变异,连续分子学监测对该地区的疫苗接种政策有重要意义。
Objective To analyze the characteristics of the HA1 gene of influenza B virus strains in Tangshan during October2011-March 2016. Methods The throat swab specimens from patients displaying symptom of influenza-like illness in sentinel surveillance hospital were conducted for the test of nucleic acid. The positive samples were isolated using MDCK cells culture. And HA1 genome of 10 isolates of influenza B virus were sequenced,so as to analyze the characteristics of nucleic acid sequence and amino acid variations. Results Phylogenetic analysis showed that the influenza B virus were Victoria lineage and Yamagata lineage,in which 4 strains of B/Victoria belong to clade 1B( 2011-2012) and clade 1A( 2015-2016),and 6 stains of B/Yamagata belong to clade 3. Compared with vaccine strain B/Brisbane/60/2008,the amino-acid substitutions occurred in N129 S and I146V( clade 1B),and I117 V and N129D( clade 1A). Compared with vaccine strain B/Wisconsin/1/2010( B/Yamagata clade 3),the amino acid at 7 positions were replaced. Compared with vaccine strain B/Massachusetts/2/2012( B/Yamagata clade 2),B/Y strains showed changes at 14 HA positions,four of which( N116 K,S150I,N165 Y,D196N) were located in four antigenic sites. Conclusion Significant variation was found among influenza B strains isolated in Tangshan area from 2011 to 2016,and it's important to carry out continuous genetic surveillance for better vaccination policy in this region.
