关附甲素对心肌细胞电生理特性的影响 被引量:23

Effects of Guanfu Base A on Electrophysiological Properties of Guinea Pig Myocardial Cells

摘要 用细胞内微电极术结合微机自动分析,研究关附甲素(GFA)对豚鼠乳头状肌快反应动作电位的影响。结果表明GFA(2.6~260μmol/L)可依浓度降低APA和V_(max),增加V_(MT),且使V_(max)恒定出现在膜电位除极至-27+SD6 mV水平。GFA(2.6~26μmol/L)使APD_(50)、APD_(90)和ERP延长,82~260μmol/L时,APD_(50)、APD_(90)缩短,而ERP/APD_(90)增大。GFA对V_(max)的抑制作用呈明显的频率依赖性和电位依赖性,并使心肌细胞的兴奋性降低。表明GFA主要是通过对快Na通道阻滞而发挥其抗心律失常作用的。 Guanfu base A is a new alkaloid first isolated from the tuber of Aconitum coreanum in China. The electrophysiological effects of guanfu base A were investigated on the isolated papillary muscles of guinea pigs by intracellular microelectrode technique coupled with microcomputer analysis. Guanfu base A (2.6~260 μmol/L) decreased APA and V_(max), increased the ratio (V_(mt)) which the time from the beginning of fast depolarization to V_(max) divided by the time of phase 0, in concentration-dependent manner. But V_(max) always occured when the membrane potential reached—27+6 mV levels, and had less effect on RP. Guanfu base A (2.6~260 μmol/L) prolonged APD and ERP, at high concentration (82~260 μmol/L), APD was shortened but ERP was prolonged relatively. The excitability of papillary muscle was reduced by guanfu base A (26 μmol/L). The inhibition of guanfu base A on V_(max) showed frequency dependent and potential dependent effects. The above results suggested that guanfu base A could block the fast Na channels and exhibited anti-arrhythmic action.
