丹参酮ⅡA磺酸钠在百草枯中毒中的心肌保护作用 被引量:2

摘要 目的观察百草枯中毒鼠心肌细胞中Bcl-2/Bax的表达,同时探讨丹参酮ⅡA磺酸钠心肌保护机制。方法 120只健康成年SD大鼠随机分为3组,每组40只。PQ染毒组大鼠PQ 80mg/kg一次性灌胃染毒,对照组等量无菌生理盐水灌胃。丹参酮干预组于PQ 80mg/kg一次性灌胃染毒后30min内给予丹参酮IIA磺酸钠注射液1ml/kg·d进行腹腔注射,同时给予对照组和PQ染毒大鼠腹腔注射等量生理盐水。3组分别于染毒后12h、24h、48h、72h各处死10只大鼠,取心肌组织处理后分别采用Western blot法和RT-PRC法检测检测心肌促凋亡基因(Bax)以及抑制凋亡基因(Bcl-2)的蛋白水平和m RNA水平。结果 PQ 12h,24h,48h和72h各组大鼠Bcl-2蛋白表达与对照组相比明显降低,而Bax蛋白表达明显升高(P<0.05或0.01);在STS24h,48h和72h组心肌细胞Bcl-2蛋白表达明显高于同期PQ各组(P<0.05或0.01),而12h,24h,48h和72h组Bax蛋白表达均明显低于同期PQ各组(P<0.05或0.01);PQ12h,24h,48h和72h各组与对照组比较心肌细胞Bax m RNA表达明显增加,Bcl-2 m RNA表达明显下降(P<0.05或0.01);与PQ染毒组比较,STS48h和72h组心肌细胞Bax m RNA表达明显降低,Bcl-2 m RNA表达明显增加(P<0.05或0.01)。结论丹参酮ⅡA磺酸钠可能通过调节Bcl-2/Bax凋亡相关基因的表达,减轻百草枯中毒大鼠心肌细胞的凋亡,发挥心肌保护作用。 Objective To observe the expression of Bcl-2/Bax in myocardial cells of the paraquat-poisoning rats and its relationship with the protection of tanshinoneⅡA sodium sulfonate. Methods 120 healthy adult SD rats were randomly divided into 3groups(n=40). The rats in PQ group were given a single intragastric administration of PQ(80mg/kg). The rats in control group were given with equal amount of saline. The rats in STS group were given a single intraperitoneal injection of STS(1ml/kg/d) at 30 min following PQ exposure. Rats in control and PQ groups were intraperitoneally injected with equal amount of saline. At 12,24,48,and 72 h after PQ exposure,10 randomly selected rats from each group were anesthetized and sacrificed by abdominal aorta bloodletting. The expression of Bax and Bcl-2 in the myocardial cells were detected by Western blot. The expression of Bax and Bcl-2m RNA in the myocardial cells were measured by RT-PCR. Results Compared with control group,the Bcl-2 protein expressions and m RNA expressions in PQ groups exhibited significantly lower,but notably higher Bax protein expressions and m RNA expressions at 12,24,48 and 72 h after PQ exposure(P〈0.05 or 0.01). Bcl-2 protein expression in STS 24 h,48h and 72 h group were significantly higher,but Bax expression in STS 12 h,24h,48 h and 72 h group were lower compared with PQ group at the same time point(P〈0.05 or 0.01). In STS48 h and 72 h group,the Bcl-2 m RNA expressions were higher and Bax m RNA expressions were lower respectively compared with PQ group(P〈0.05or0.01). Conclusion Tanshinone Ⅱ A sulfonate may reduce the myocardial cell apoptosis of paraquat-poisoning rats by adjusting the Bcl-2/Bax apoptosis related gene expression.
