摘要
以夏天无块茎为试材,采用不同浓度秋水仙素(0.00%、0.05%、0.10%、0.20%)添加2%二甲基亚砜(DMSO)分别浸泡24、48、72h的方法,研究了不同浓度秋水仙素和不同处理时间对染色体加倍的影响。结果表明:以0.20%秋水仙素处理24h较优,成活率为31.4%,加倍率为50.0%,其次为0.10%秋水仙素处理24、48h和0.20%秋水仙素处理48h。其中,未加倍的植株染色体数为2n=28,加倍后的植株为2n=56。
Tubers fromCorydalis decumbens(Thunb.)Pers.were immersed in 0.00%,0.05%,0.10%and 0.20% colchicine solution with 2% DMSO for 24 hours,48hours,72 hours,respectively.The effects of different concentrations of colchicine and different treatment time on chromosome doubling were studied.The results indicated that the better treatment was 0.20% colchicine solution for24 hours with a doubling rate of 50.0% and a survival rate of 31.4%.Secondly for 0.10% colchicine solution for 24 hours,48hours and 0.20% colchicine solution for 48 hours.The chromosome number of diploid Corydalis decumbens(Thunb.)Pers.was 2n=28and that of tetraploid was 2n=56.
