适于新疆制干类型辣椒种质资源遗传多样性的SRAP和SCoT分析 被引量:15

摘要 本研究对72份制干辣椒种质资源采用SRAP和SCoT分子标记技术进行遗传多样性研究。以期从分子水平上揭示新疆制干辣椒种质资源间的亲缘关系,为其种质搜集、利用和遗传改良提供一定的理论基础。结果表明,在10对SRAP引物组合共检测出78个位点,其中多态性位点数69个,多态性比例为88.64%,平均每条引物扩增位点为7.8个,PIC为0.819;20对SCoT引物扩增出203个位点,其中多态性位点数174个,多态性比例为85.71%,平均每条引物扩增位点为10.15个,PIC为0.861。UPGMA聚类结果显示,在相似性系数为0.74时,72份材料可被分为2个类群;阈值为0.79时类群2又可分为5组,表明制干辣椒种质间具有相对丰富的遗传多样性。分析结果将有助于更好地保护和利用制干辣椒种质资源,研究结果为进一步进行制干辣椒的种质研究及利用提供了帮助。 In order to reveal the genetic relationship of pepper germplasm at molecular level and provide some the oretical basis for germplasm collection, identification, utilization, genetic improvement, 72 genotypes of dried-orientation pepper germplasm was employed in this study and the genetic diversity was analyzed by SRAP and SCo T molecular marker. The results showed that there are 78 locis were found by 10 SRAP primer pair combinations and 69 of these locis were polymorphic locis. The polymorphism ratio was 88.64% and averagely7.8 locis could be amplified by each SRAP primer, while the PIC was 0.819. Furthermore, 20 SCo T primer pairs amplified 203 sites which had 174 polymorphic locis and the polymorphism ratio was 85.71%. Averagely, 10.15 locis could be amplified by each SCo T primer and the PIC is 0.861. The result of UPGMA cluster analysis showed that 72 pepper germplasm could be divided into two groups when the similarity coefficient was at 0.74, while the second group could be divided into five sub groups when the threshold was at 0.792, which indicated that dried-orientation pepper had a relatively rich genetic germplasm diversity. The result of this study help to protect and utilize the dried-orientation pepper germplasm, moreover, the result could be helpful for further improvement and utilization of dried-orientation pepper germplasm.
