柿叶黄酮治疗实验性口腔溃疡的作用及其抗炎机制研究 被引量:3

Study on the effect and the anti-inflammatory mechanisms of persimmon leaf flavonoid in the treatment of experimental oral ulcer

摘要 目的探讨柿叶黄酮对大鼠口腔溃疡的治疗作用及其抗炎机制。方法 60只SD大鼠,分为空白对照组、模型对照组、阳性对照组及柿叶黄酮20、40、80 mg/kg组,每组10只。空白对照组不做处理,其他各组建立口腔溃疡大鼠模型(醋酸法),空白对照组、模型对照组给予灌胃等体积生理盐水,阳性对照组给予灌胃西瓜霜,柿叶黄酮组给予灌胃相应剂量柿叶黄酮。比较造模后第4、7天的溃疡面积,测定溃疡组织中肿瘤坏死因子-α(TNF-α)水平。结果用药后4、7 d,阳性对照组、柿叶黄酮40 mg/kg组、柿叶黄酮80 mg/kg组大鼠的溃疡面积均小于模型对照组,差异具有统计学意义(P<0.05)。模型对照组(66.5±4.1)ng/L、阳性对照组(38.7±4.7)ng/L、柿叶黄酮组[(40.2±3.8)、(35.8±4.6)、(32.6±3.8)ng/L]大鼠溃疡组织内TNF-α水平高于空白对照组(27.8±3.2)ng/L,差异具有统计学意义(P<0.05)。阳性对照组、柿叶黄酮组大鼠溃疡组织内TNF-α水平低于模型对照组,差异具有统计学意义(P<0.05)。结论柿叶黄酮对醋酸诱导的口腔溃疡有一定的治疗作用,其机制可能与降低炎症反应有关。 Objective To investigate the therapeutic effect of persimmon leaf flavonoid on oral ulcer in rats and its anti - inflammatory mechanism. Methods A total of 60 SD rats were divided into blank control group,model control group and positive control group, persimmon leaf flavonoid 20, 40 and 80 mg/kg group, with 10 rats in each group. The blank control group received no treatment, and the other groups established the models of rat oral ulcer (acetic acid). Blank control group, model control group was treated with intragastric administration of equal volume normal saline, the positive control group was treated with intragastrie administration of watermelon frost, and persimmon leaf flavonoid group was treated with intragastric administration of corresponding dose persimmon leaf flavonoid. The ulcer area was compared after 4 and 7 days of modeling, and the level of tumor necrosis factor- α (TNF- α ) in ulcer tissue was measured. Results After 4 and 7 d of medication, the positive control group, persimmon leaf flavonoid 40 and 80 mg/kg group had smaller ulcer area than model control group, and the difference had stat!stical significance (P〈0.05). The model group (66.5± 4.1) ng/L, positive control group (38.7 ± 4.7) ng/L and persimmon leaf flavonoid group [ (40.2 ± 3.8), (35.8 ± 4.6) and (32.6 ± 3.8) ng/L ] had higher TNF- α level in ulcer tissue than bland control group (27.8 ± 3.2)ng/L, and their difference had statistical significance (P〈-.05). The positive control group and persimmon leaf flavonoid group had lower TNF- ot level in ulcer tissue than model control group, and the difference had statistical significance (P〈0.05). Conclusion Persimmon leaf flavonoid have a certain therapeutic effect on acetic acid-induced oral ulcers, and its mechanism may be related to the reduction of inflammatory response.
