不同艾灸预处理时间对心肌缺血再灌注损伤大鼠心肌细胞中HIF-1α、Bcl-2及Caspase-3表达的影响 被引量:16

Effect of Different Length of Moxibustion Preconditioning on the Expression of HIF-1α,Bcl-2 and Caspase-3 in Myocardial Cells of MIRI Rats

摘要 目的:通过观察不同艾灸预处理时间对心肌缺血再灌注损伤大鼠心肌细胞中低氧诱导因子-1α(HIF-1α)、B细胞淋巴瘤/白血病-2(Bcl-2)及天冬氨酸特异性半胱氨酸蛋白酶-3(Caspase-3)蛋白和基因表达的变化,探讨艾灸预处理对MIRI预防保护机制。方法:将40只大鼠分为正常组、模型组、艾灸预处理5天组、艾灸预处理10天组。取"内关"穴行艾灸预处理5 d和10 d后,采用冠脉结扎法建立MIRI模型。运用实时荧光定量PCR(Real-time PCR)技术以及Western blot方法检测大鼠心肌细胞中Bcl-2、HIF-1α及Caspase-3蛋白及基因表达。结果:与正常组比较,模型组HIF-1α和Caspase-3蛋白及mRNA表达均高于正常水平(P<0.01),Bcl-2蛋白及mRNA表达低于正常水平(P<0.01)。与模型组比较,艾灸预处理组HIF-1α蛋白及mRNA表达降低(P<0.01),Bcl-2蛋白及mRNA表达增高(P<0.01),Caspase-3蛋白及mRNA表达下降(P<0.01 or P<0.05)。与5天组比较,艾灸预处理10天组HIF-1αmRNA表达明显降低(P<0.05),Bcl-2蛋白显著增高(P<0.05),Caspase-3蛋白及mRNA表达显著下降(P<0.01)。结论:艾灸预处理可通过降低MIRI大鼠心肌细胞中HIF-1α、Caspase-3的含量,增加心肌细胞中Bcl-2的含量,改善心肌缺血缺氧状态以及抑制心肌细胞凋亡,对损伤心肌细胞起到预防保护作用。其中艾灸预处理10天对心肌细胞保护作用优于5天。 Objective: To discuss the protective mechanism of moxibustion preconditioning for myocardial ischemia reperfusion injury( MIRI),by observing the expression changes of HIF-1α,Bcl-2,and Caspase-3 protein and mRNA in myocardial cells of MIRI rats by different length of moxibustion preconditioning. Methods: 40 rats were randomly divided into the normal group,the model group,the moxibustion preconditioning group A( preconditioning for 5 days),and moxibustion preconditioning group B( preconditioning for 10 days). Moxibustion preconditioning was applied on point of PC6 for 5 or 10 days respectively before MIRI model being established by coronary artery ligation. The expressions of HIF-1α,and Caspase-3 protein and mRNA in myocardial cells were detected with RT-PCR technique and Western blot method. Results: The protein and mRNA expressions of HIF-1α,and Caspase-3 in the model group was higher than the normal level( P〈0. 01); the protein and mRNA expressions of Bcl-2 in the model group was lower than the normal level( P〈0. 01). Compared to the model group,the expressions of HIF-1α and Caspase-3 in the moxibustion preconditioning groups decreased significantly( P〈0. 05,P〈0. 01),the expressions of Bcl-2 increased significantly( P〈0. 01),of which moxibustion preconditioning group B was superior to group A( P〈0. 05,P〈0. 01). Conclusion: Moxibustion preconditioning can decrease the expression of HIF-1α and Caspase-3 and increase the expression of Bcl-2 in myocardial cells to improve myocardial ischemia and inhibit myocardial apoptosis. The protective effect of moxibustion preconditioning for 10 days was better than that for 5 days.
