敦煌古方小补心汤对大鼠微血管内皮细胞的保护作用研究

摘要 目的探讨《辅行诀脏腑用药法要》中记载的敦煌古方小补心汤对高糖条件下大鼠心肌微血管内皮细胞(MMVECs)的保护作用。方法采用敦煌古方小补心汤含药鼠血清干预高糖培养条件下的MMVECs,流式细胞技术观察MMVECs的增殖活力,ELISA法检测MMVECs培养基中血管内皮生长因子(VEGF)分泌量、Q-PCR法检测MMVECs中VEGF的基因表达水平。结果小补心汤能促进MMVECs增殖,上调MMVECs的VEGF基因表达,增加VEGF分泌。结论敦煌古方小补心汤在一定程度上能改善糖尿病引起的心血管损伤。 Objective To explore the protective effects of Dunhuang ancient Xiaobuxin Decoction,recorded in "Auxiliary Drug-use Regulations for Zang-Fu",on myocardial microvascular endothelial cells(MMVECs) in rats under the condition of high glucose.Methods MMVECs intervened with Dunhuang ancient Xiaobuxin Decoction medicated serum and cultured under the condition of high glucose were observed,and flow cytometry was used to detect the proliferation activity of MMVECs,ELISA to detect the medium(VEGF) secretion of MMVECs cultured vascular endothelial growth factor,Q-PCR to detect VEGF gene expression level of MMVECs.Results Xiaobuxin Decoction could promote MMVECs proliferation,increase the expression of VEGF gene of MMVECs,and increase the secretion of VEGF.Conclusion Dunhuang ancient Xiaobuxin Decoction could relief cardiovascular injury caused by diabetes to some extent.
