葡萄糖及胰岛素对牛血管内皮细胞凋亡的影响 被引量:38

Effect of different concentrations of glucose and insulin on apoptosis of bovine vascular endothelial cells

摘要 目的通过对牛血管内皮细胞(EC)的体外培养研究糖尿病(DM)状况下葡萄糖(Glu)、胰岛素(Ins)对EC凋亡的影响。方法吖啶橙/溴乙锭(AO/EB)荧光染色及DNA片段的琼脂糖凝胶电泳对EC凋亡定性,ELISA方法测定DNA片段定量EC凋亡。结果高浓度Glu(20mmol/L、40mmol/L)和Ins(300mU/L、3000mU/L)能够诱发EC发生凋亡。EC凋亡与Glu和Ins浓度成正比,并且为时间依赖性。低浓度Ins(30mU/L)明显改善Glu40mmol/L下所致EC的凋亡,但高浓度Ins与Glu有正性协同作用,增加EC凋亡。高浓度甘露醇(Man20mmol/L、40mmol/L)不引起EC明显凋亡。结论DM环境下高血糖、高Ins血症能诱发EC凋亡,促进DM血管并发症的发生。 Objective To study the effects of different concentrations of glucose and insulin on apoptosis of cultured endothelial cells (ECs). Methods Apoptasis of ECs was studied qualitatively with acridine orange(AO)/ethidium bromide(EB) fluorescence staining and DNA agarose gel electrophoresis and quantitatively with ELISA method. Results High concentrations of glucose and insulin induced apoptosis. Apoptosis of ECs was positively correlated with concentrations of glucose and insulin and was time dependent. Low concentration of insulin decreased apoptosis of ECs induced by glucose but high concentrations of insulin had synergistic action with glucose and increased apoptosis of ECs. High concentrations of mannitol did not induce obvious apoptosis of ECs. Conclusion High plasma glucose and insulin levels in diabetes mellitus may induce opoptosis of vascular endothelium and accelerate the development of vascular complications.
