野生蒲公英花多酚的提取和体外抗氧化活性研究 被引量:13

Extraction and Antioxidant Activity in vitro of Polyphenols from Northeast Wild Dandelion Flowers(Toraxacum mongolicum Hand.Mazz)

摘要 以Fenlin-Ciocalten法测定蒲公英花多酚(DFP)含量,通过单因素和正交实验来确定DFP的最佳提取条件,并对其体外抗氧化活性进行了研究。实验结果表明,最佳提取工艺为:乙醇体积分数40%,料液比1∶15(g·m L^(-1)),提取温度40℃,提取时间40 min。在最优提取条件下DFP的得率为2.31%。抗氧化试验结果显示,在一定浓度范围内DFP对1,1-二苯基-2-三硝基苯肼(DPPH)、羟自由基(·OH)、超氧阴离子自由基(O_2^-·)的清除率分别为68.43%、89.35%、76.55%,表现出良好的抗氧化能力。表明东北野生蒲公英可以作为功能性食品原料具有良好的开发前景。 The optimization extraction of total polyphenols from northeast wild dandelion flowers was studied by single factor test and orthogonal test. Results showed that the optimal conditions were 40% ethanol solvent,extraction temperature 40℃, 1:15 ratio of material to ethanol,extraction time 40 min. Under the extraction conditions,the extraction rate of total polyphenols was 2.31%. Antioxidant activities in vitro of the dandelion flower polyphenols (DFP)were investigated in three different assays,including scavenging activities of hydroxyl free radicals (·OH),superoxide anion (O2-·)and 1,1-Diphenyl-2-picrylhydrazyl radical 2,2- Diphenyl-1- (2,4,6-trinitrophenyl)hydrazyl (DPPH)free radicals. Results showed that the highest scavenging rates for the radicals were 89.35% ,76.55% and 68.43% ,respectively. As a conclusion,DFP had good eliminate ability of free radical,could be used as functional food material.
