摘要
目的探讨β-arrestin 2的小分子干扰RNA(small interfering RNA,siRNA)对人乳腺癌Bcap37细胞增殖的影响。方法在脂质体Lipofectamine^(TM) 2000的介导下用β-arrestin 2 siRNA转染Bcap37细胞,同时设置空白对照组(不作任何处理)及阴性对照组(转染p GPU6/GFP/Neo SHNC)。转染后用实时荧光定量PCR(quantitative realtime polymerase chain reaction,qRT-PCR)及Western blot法检测Bcap37细胞β-arrestin 2基因及蛋白的表达情况,MTT法检测Bcap37细胞的增殖。结果转染96 h后β-arrestin 2 siRNA转染的Bcap37细胞中β-arrestin 2的基因及蛋白表达低于空白及阴性对照组(均P<0.05),并且转染96 h后β-arrestin 2 siRNA转染的细胞增殖速度低于空白及阴性对照组(均P<0.05)。结论β-arrestin 2 siRNA降低Bcap37细胞β-arrestin 2的表达并抑制细胞增殖。
Objective To investigate the effect of β-arrestin 2 small interfering RNA( siRNA) on the proliferation of human breast cancer Bcap37 cells. Methods Bcap37 cells were transfected with β-arrestin 2 siRNA by lipofectamineTM2000. p GPU6/GFP/Neo SHNC transfected cells were used as the negative control and the non-transfected cells served as the blank control. The expression of β-arrestin 2 gene was analyzed by quantitative real-time polymerase chain reaction( qRT-PCR) and Western blot. Cell proliferation was examined by MTT assay. Results The expression level of β-arrestin2 was significantly lower in the β-arrestin 2 siRNA group than that in the negative control or blank control groups( both P〈0. 05). The cell proliferation rate of β-arrestin 2 siRNA transfected cells was also significantly lower than that of two control groups( both P〈0. 05). Conclusion β-arrestin 2 siRNA can effectively down-regulate the expression of β-arrestin 2,and inhibit the proliferation of human breast cancer Bcap37 cells.
