脊髓损伤新生小鼠脑源性神经营养因子前体对少突胶质细胞存活及生长的影响 被引量:2

摘要 目的 研究脊髓损伤新生小鼠脑源性神经营养因子前体对少突胶质细胞存活及生长的影响.方法 分离新生小鼠大脑皮层少突胶质细胞前体细胞,进行体外原代培养;采用WB法观察脑源性神经营养因子前体及其受体分拣蛋白与神经营养素受体的表达水平.将浓度为1 μg·L-1脑源性神经营养因子前体设置为A组,10 μg·L-1脑源性神经营养因子前体设置为B组,100 μg·L-1脑源性神经营养因子前体设置为C组,空白对照组为100 μg·L-1牛血清白蛋白;每组设置6个平行副孔.采用β-tublin和结晶紫进行染色,观察少突胶质细胞前体细胞的细胞形态,评估不同浓度的脑源性神经营养因子前体对少突胶质细胞前体细胞的生产情况的影响.结果 脑源性神经营养因子前体及其受体分拣蛋白与神经营养素受体在少突胶质细胞前体细胞中的表达均呈现阳性;A组、B组、C组中的少突胶质细胞前体细胞的数量显著低于对照组,且随着浓度的增加,少突胶质细胞前体细胞数量逐渐下降,各组间比较差异均有统计学意义(P〈0.05);与对照组比较,A组、B组、C组少突胶质细胞前体细胞突起显著减少,且面积显著缩小,各组间比较差异有统计学意义(P〈0.05).结论 少突胶质细胞前体细胞可以内源性的表达脑源性神经营养因子前体F以及其受体,而脑源性神经营养因子前体的浓度对少突胶质细胞前体细胞的生长和存活具有显著的抑制作用,且这一作用机制可能与神经营养素受体-分拣蛋白通路相关. Objective To investigate the influence of proBDNF of neonatal mouse with spinal cord injury on survival and growth of oligodendroglia cell. Methods Cerebral cortex OPCs of neonatal mouse was separated and primarily cultured; expression level of proBDNF and its receptor sortilin and neurotrophins receptors were observed by WB. proBDNF with concentration of 1 μg · L-1 , 10 μg · L-1 , 100 μg · L-1 were respectively seen as group A, B, C. 100 μg · L -1 BSA were seen as blank control group; 6 parallel holes were set in each group. β-tublin and crystal violet were used for staining, cellular morphology of OPCs was observed, influence of proBDNF with different concentrations on production status of OPCs was evaluated. Results Expression of proBDNF and its receptor sortilin and neurotrophins receptors in OPCs was positive; the number of OPCs in group A, B, C was obviously lower than control group, and with the increasing of concentration, the number of OPCs was gradually declined, the difference among groups was statistic significant (P〈0.05). Compared with control group, OPCs of group A, B, C were significantly declined and the area was shrinked, the difference among groups was statistic significant (P〈0.05). Conclusion OPCs can endogenously express proBDNF and its receptor, and the concentration of proBDNF has a significant inhibitory effect on the growth andsurvival of OPCs, and this mechanism may be related to neurotrophins receptors-sortilin pathway.
