摘要
高脂血症和肥胖已成为严重的公共健康问题,引起了人们的广泛关注。脂肪变性细胞模型是研究降脂减肥的重要手段之一。比较分析了诱导人肝L-02细胞、人肝HepG2细胞、小鼠3T3-L1前脂肪细胞、大鼠肝BRL细胞、人肝Changliver细胞、奶牛肝细胞、草鱼肝细胞(L8824)等不同细胞株建立高脂细胞模型的方法和效果。诱导多在细胞对数生长末期,并多采用脂肪酸、脂肪乳、高浓度胎牛血清等诱导培养24~48 h。不同细胞株及不同的诱导方法,细胞中甘油三酯(TG)和总胆固醇(TC)的升高幅度有较大差异。
Hyperlipidemia and obesity become serious public health problems,attracting widespread attention.Cell model is one of the important means of study lipid-lowering diet.This paper compared and analyzed the methods and effect of different types of cell lines to set up high fat cell model,including human liver L-02 cells,human HepG2 liver cells,3T3-L1 preadipocytes in mice,rat liver BRL cells,human liver Changliver cells,cows liver cells and grass carp liver cells(L8824).Most of inducing high fat cells were in the end of logarithmic phase,induced by fatty acid,fat emulsion and high concentrations of fetal bovine serum,the induction time about 24 to 48 hours.Different cell lines and induction method lead to large different ascending range in triglycerides(TG) and total cholesterol(TC).
