诱导型一氧化氮合酶(iNOS)对重症急性胰腺炎时胰岛内分泌功能的影响 被引量:2

摘要 目的研究重症急性胰腺炎大鼠胰腺内分泌功能的变化,探讨iNOS在胰腺内分泌功能损伤中的作用。方法2015年7月—2016年7月在济宁医学院动物实验中心取健康雄性Wistar大鼠,体重250~300 g,24只随机分成2组,对照组12只,SAP组12只,应用5%牛磺胆酸钠逆行注入大鼠胰胆管制作重症急性胰腺炎(severe acute pancreatitis,SAP)大鼠模型,检测血清淀粉酶、血糖浓度;分离胰岛,行葡萄糖刺激胰岛素分泌试验;应用NOS试剂盒检测胰岛iNOS的活性。结果 SAP组血糖、血清淀粉酶、葡萄糖葡萄糖刺激胰岛素分泌实验和胰岛的iNOS活性分别为:(11.50±1.36)mmol/L、(1 575.36±435.33)m IU/L、(3.90±0.78)m IU/L、(10.36±2.14)m IU/L和(30.27±3.57)U/m L;对照组分别为:(5.32±1.25)mmol/L、(104.18±35.58)m IU/L、(7.82±1.03)mmol/L、(25.60±3.72)mmol/L和(7.79±0.58)U/m L;两组分别相比差异有统计学意义(P<0.05)。结论重症急性胰腺炎时对胰岛功能影响较大SAP胰岛功能损伤与iNOS活性增强有关系,对胰岛素分泌产生负面影响。 Objective To research the changes of rat endocrine function of severe acute pancreatitis and study the effect of iNOS in the pancreatic endocrine function injuries. Methods 24 healthy male rats in the animal experimental center of Jining Medical College whose weight was 250-300 g from July 2015 to July 2016 were randomly divided into two groups with 12 rats in each, and the serum amylase and blood glucose concentration were tested and the insulin was isolated and the rats were given the glucose-stimulated insulin secretion test, and the insulin iNOS activity was tested by the NOS kit.Results The differences in the blood glucose, serum amylase, glucose and glucose stimulated insulin secretion test and iNOS activity of insulin between the SAP group and the control group were statistically significant [(11.50 ±1.36)mmol/L,(1 575.36±435.33)mIU/L,(3.90±0.78)mIU/L,(10.36±2.14)mIU/L,(30.27±3.57) U/mL vs(5.32±1.25)mmol/L,(104.18±35.58)mIU/L,(7.82±1.03)mmol/L,(25.60±3.72)mmol/L(7.79±0.58)U/mL](P〈0.05). Conclusion The effect of severe acute pancreatitis on the insulin function is bigger, and the SAP insulin function injury is related to the enhancement of iNOS activity and has a negative effect on the insulin secretion.
