氧化型低密度脂蛋白对血管平滑肌细胞应激活化蛋白激酶活性及凋亡的影响

摘要 目的 观察氧化型低密度脂蛋白 (Ox LDL)对血管平滑肌细胞凋亡及应激活化蛋白激酶 (SAPK)活性的影响。方法 采用 3个时间水平 (2 4、48、72h)、4个剂量水平 (0、 5 0、 10 0、 2 0 0μg/ml)的两因素析因设计观察Ox LDL对兔主动脉血管平滑肌细胞 (vascularsmoothmusclecells ,VSMCs)凋亡影响的时间、剂量效应关系 ;Ox LDL对兔主动脉血管平滑肌细胞SAPK活性影响。结果 Ox LDL诱导VSMCs凋亡随时间和剂量升高而不断增加 ,呈现一定的时间、剂量效应关系 (P <0 0 1) ;Ox LDL可引起各血管平滑肌细胞SAPK活性升高。结论 Ox LDL诱导血管平滑肌细胞凋亡与其作用浓度、时间相关 ;SAPK信号通路可能在Ox Objective To study the effects of oxidized low density lipoprotein(Ox-LDL) on vascular smooth muscle cells VSMCs and stress-activated protein kinase (SAPK). Methods Two-factorial experiment was designed to explore the influence of four dosage levels of Ox-LDL (0 μg/ml, 50 μg/ml, 100 μg/ml, 200 μg/ml) at three different periods of exposure (24 h, 48 h and 72 h) on apoptosis of arotic vascular smooth muscle cells of rabbit. Activities of SAPK in the cultural vascular smooth muscle cells were observed at different exposure levels of Ox-LDL at different time. Results The apoptosis of VSMCs increased with both Ox-LDL dosage and the exposure time (P<0.01). The SAPK activity of VSMCs exposed to Ox-LDL (50 μg/ml, 100 μg/ml, 200 μg/ml) at 10 min was higher than that of the control group. Conclusion Ox-LDL induced the apoptosis of VSMCs, which were associated with exposure dose and period. The signal pathway of SAPK may play an important role in the apoptosis of VSMCs induced by Ox-LDL.
