分流型肺动脉高压模型大鼠存活素的变化 被引量:1

摘要 目的观察凋亡抑制因子存活素在分流型肺动脉高压中的变化。方法 20只wista大鼠随机分为实验组和对照组。采用左颈总动脉与左侧颈外静脉吻合法建立分流型肺动脉高压模型。右心导管法检测平均肺动脉压,测定右心室肥厚指数(右心室组织重量/左心室+室间隔组织重量),HE染色及免疫组化观察肺组织,ELISA法测定大鼠肺组织匀浆存活素和IL-6浓度。结果实验组大鼠平均肺动脉压、存活素、IL-6含量明显高于对照组,P<0.05。实验组大鼠肺小动脉管腔缩窄,管壁增厚明显。血管内皮细胞水肿。肺小动脉管周大量炎性细胞浸润,以淋巴细胞为主。肺泡间隔增宽明显。存活素在实验对照组大鼠肺组织中表达强烈,发光细胞分布于肺泡间膈中和肺动脉平滑肌中。IL-6蓝染颗粒分布于肺泡间隔中和肺动脉平滑肌中。结论凋亡因子存活素在分流型肺动脉高压发病中起到重要作用,上游细胞因子IL-6参与了存活素的激活。 Objective To observe the changes of the apoptosis inhibitor survivin in shunt type pulmonary arterial hypertension rat model. Methods 20 wista rats were randomly divided into experimental group and control group. Shunt type pulmonary hypertension model was established by anastomosis of left common carotid artery and the left external carotid vein. Mean pulmonary artery pressure be determinated by right cardiac catheterization. Right ventricular hypertrophy index (right ventricular weight/left ventricular & ventricular septal tissue weight), lung tissues were observed, by HE staining and immunohistochemical staining, ELISA method for determination concentration of rat lung survivin and of IL-6. Results The mean pulmonary artery pressure of experimental group rats, survivin, IL-6 content were significantly higher than the control group, P〈0.05. Experimental rat pulmonary arteriolar lumen narrowing, wall thickening and hyaline degeneration. Vascular endothelial cell edema. Pulmonary arteriole around full of inflammatory cells. Immunohistochemistrty showed expression of surviving and IL-6 obviously on the lung tissue in the experimental group. Conclusion Survivin apoptosis factors played an important role in pulmonary hypertension incidence of shunt shunt type pulmonary arterial hypertension rat model, and upstream cytokine IL-6 was involved in the activation of survivin.
