透析用水处理系统可培养和不可培养细菌的分布规律 被引量:3

Distribution of culturable and non-culturable bacteria in hemodialysis water treatment system

摘要 目的研究典型透析用水处理系统中可培养和不可培养细菌的分布规律,以准确评价透析用水的微生物学安全性。方法以平板计数法和死/活菌荧光染色结合激光共聚焦扫描显微镜(CLSM)观察法,分别测定可培养及不可培养细菌(包括未培养细菌和VBNC细菌)的数量,并结合工艺运行物理化学指标进行分析。结果可培养细菌水平符合相关标准,但透析用水中的水平高于自来水;不同样品中不可培养细菌数量高于可培养细菌2~6个数量级,在透析用水中约为10~4个细胞/100ml。结论培养法不能准确反映透析用水处理系统中细菌数量的分布,大部分细菌以不可培养状态存在,对透析患者的健康存在潜在威胁。 OBJECTIVE To investigate the distribution of culturable and non-culturable bacteria in hemodialysis water treatment system so as to accurately evaluate the microbiological safety of hemodialysis water.METHODS The counts of the culturable and non-culturable bacteria (including uncultured bacteria and VBNC bacteria) were determined by means of plate count method and dead or viable bacteria fluorescent staining combined with confocal laser scanning microscopy (CLSM),and the physical and chemical indexes were analyzed via process operation.RESULTS The level of culturable bacteria met the related standards,however,the level of culturable bacteria was lower in the hemodialysis water than in the running water.The counts of non-culturable bacteria in different samples were 2~6 order of magnitudes higher those of the culturable bacteria,and the counts of non-culturable bacteria were about 104 cells/100 ml.CONCLUSION The culture method can not accurately reflect the distribution of bacterial colony counts in the hemodialysis water treatment system,most of the bacteria prevail in non-culturable state,which poses potential threat to the health of the patients.
