摘要
以无籽刺梨(Rosa sterilis S.D.Shi)茎段为外植体,采用75%酒精+升汞4min40s的消毒组合,可获得63.63%无菌株得率;最佳诱导培养基为改良MS+6-BA4.0mg/L+NAA0.01mg/L;最佳增殖培养基为改良MS+6-BA1.0mg/L+NAA0.1mg/L,增殖系数2.67;最佳生根培养基为改良1/2MS+IBA0.2mg/L,生根率87.5%;采用80%腐殖土+15%珍珠岩+5%河沙的炼苗基质,前期注意保持85%的炼苗湿度,无籽刺梨组培苗成活率达90%以上。
Stem segments of seedless Rosa sterilis were used as explants, using 75 % alcohol + mercuric chloride 4 min40s disinfection combination, 63.63% of sterile plants could be obtained. The results showed that the suitable medium of ini- tial induction was improved MS medium with 4. 0 mg/L 6-BA and 0. 01 mg/L NAA; the best proliferation occurred on improved MS medium with 1.0 mg/L 6-BA and 0. 1 mg/L NAA and the multiplication coefficient was 2.67; the best rooting condition for plantlets was on improved 1/2MS medium with 0. 2 mg/L IBA and the rooting rate was 87.5%. U- sing the cultivation medium of 80% humus + 15% perlite + 5% sand and maintain 85% humidity in prophase, more than 90% survival rate could be obtained.
