摘要
目的探讨长链非编码RNA FEZ家族锌指1反义RNA1(FEZF1-AS1)在人胃癌组织中表达及临床意义。方法收集39例胃癌及其配对正常胃黏膜组织,采用荧光实时定量聚合酶链反应(PCR)方法检测组织中FEZF1-AS1表达水平,分析其表达与患者性别、年龄、肿瘤部位、肿瘤直径、组织分化程度、肿瘤浸润深度、淋巴结转移、血管浸润、神经浸润、肝转移数目及肝转移发生时间等临床病理因素关系,并作统计学处理。结果在39例胃癌组织中FEZF1-AS1阳性率为93.08% (36/39),而正常胃组织FEZF1-AS1阳性率为2.56% (1/39),χ2=62.986,P=0.040,差异有统计学意义。FEZF1-AS1表达量与患者性别(χ2=1.224,P=0.345)、年龄(χ2=2.377,P=0.395)、肿瘤位置(χ2=3.029,P=0.405)、组织分化程度(χ2=0.883,P=0.557)、神经浸润(χ2=0.522,P=0.470)无明显相关相关,但与原发灶直径(χ2=4.672,P=0.031)、肿瘤浸润深度(χ2=9.120,P=0.010)、淋巴结转移(χ2=8.176,P=0.014)、血管浸润(χ2=14.857,P=0.004)、肝转移数目(χ2=12.594,P=0.006)、术后肝转移发生时间早晚(χ2=7.419,P=0.038)均显著相关。结论FEZF1-AS1可能是胃癌异时性肝转移重要靶点之一。
ObjectiveTo investigate the expression and clinical significance of long chain non-coding RNA FEZ family zinc finger 1 antisense RNA 1 (FEZF1-AS1) in human gastric cancer.MethodsThirty-nine pairs of human gastric cancer and matched normal gastric tissue were collected, and FEZF1-AS1 was detected by fluorescence quantitative real-time quantitative polymerase chain reaction (Real-time PCR). The correlation between FEZF1-AS1 expression and patient’s sex, age, tumor location, tumor diameter, histological differentiation, depth of tumor invasion, lymph node metastasis, infiltration of blood vessels, nerve infiltration, the number of liver metastases, the time of liver metastasis, and distant metastasis was analyzed, respectively.ResultsThe positive rate of FEZF1-AS1 in 39 cases of gastric cancer was 93.08% (36/39), and that in normal gastric tissues was 2.56% (1/39) (χ2=62.986, P=0.040). FEZF1-AS1 expression was not significantly related with the age (χ2=1.224, P=0.345) and sex (χ2=2.377, P=0.395) of patients, tumor location (χ2=3.029, P=0.405), histological differentiation (χ2=0.883, P=0.557), and neural invasion (χ2=0.522, P=0.470), but with the primary tumor diameter (χ2=4.672, P=0.031), depth of tumor invasion (χ2=9.120, P=0.010), lymph node metastasis (χ2=8.176, P=0.014), vascular invasion (χ2=14.857, P=0.004), the number of liver metastases (χ2=12.594, P=0.006), and occurrence time of liver metastasis (χ2=7.419, P=0.038).ConclusionThese results indicate that FEZF1-AS1 is closely related to gastric cancer with metachronous liver metastasis. FEZF1-AS1 might be one of the important targets of metachronous liver metastasis in gastric cancer.
